圓管式血流流體腔設計製作與細胞培養

摘要


本論文以流體腔設計製作及細胞培養,其整體架構分為兩部分: 流體腔設計製作:利用Pro/E及Solidworks設計出體外流體腔系統圖,再利用UG繪圖軟體將所設計好之流體腔繪製並轉NC碼進行CNC加工。加工後,進行流體腔運轉測試。 細胞培養:為了證實此流體腔是可用於體外培養,先將內皮細胞培養於玻片上,再將玻片放入流體腔內;並給予符合人體內血管之剪應力。 圓管式流體腔主要分為三代設計: 1. 第一代設計,主要沿用平板式流體腔之設計理念,但發現此設計並不適用於圓管式流體腔,且製造、架設系統複雜。 2. 第二代設計和第一代設計主要不同於改良流體出入口位置,避免流體在出入口產生滯流現象,且加工容易。 3. 第三代設計和第二代設計主要不同於細胞培養段之差異,因為管璧為曲面,不像平板式流體腔為平面,所以在種植細胞之玻片有所不同,以致於改良上蓋設計。 其實驗結果顯示,圓管式流體腔成功運用於體外細胞培養系統,在剪應力 = 3pa、培養時間為六小時,成功讓內皮細胞存活,且內皮細胞從橢圓形改變成長梭形,證明本研究之體外流體腔設計是符合人體血管細胞的生長環境。

In this study, the circular flow chamber was designed and fabricated for cellculture in vitvo. The main framework which can be divided into two parts: 1st phase : Design and manufacturing of flow chamber Pro/E and Solidworks computer assist programs were introduced to design and simulate the process of manufacturing from for CAD design to motion. Therefore, Convert the results of stimulating from animation into Numerical codes that insert to Computer Numerical Control machine executed the fabrication plan. After process of machining, the experiment of the flow chamber was investigated. 2nd phase : Cell culture In order to confirm the type of flow chamber system could be in vitro, a human endothelial cell was cultured in this chamber. To modulate the shear stress as mankind for the environment of the chamber. The design of circular flow chamber are divided into three part: 1.The first generation design of flow chamber :The major concepts of this generation is basis on the parallel plate flow chamber design. However, the estimation of fabrication is found out that is hard to set up and modify for circular flow type. 2.The second generation design of flow chamber : The differentiation of 1st and 2 nd generation of flow chamber is the position of in-put and out-put. To reduce the hazard of turbulent flow on the corner near the in-put , and also simplify to be manufacturing. 3.The third generation of flow chamber: The differentiation of 2nd and 3 rd generation of flow chamber is the cell culture holder. Because the wall of tube is circular that is not horizontal as plate chamber, seeding might be on various micro-slide plate as the improvement of this design. In order to confirm the type of flow chamber system could be in vitro, a human endothelial cell was fixed on microsolide than put into cell culture chamber of this circular flow chamber model. To modulate the shear stress as 3 pascal as mankind for the environment of the chamber. The results showed that the cell can grow around the environment and the morphology of endothelial cell has transfer from oval-shaped to Spindle-shaped. According to the performance of endothelial cell results, the design of flow chamber is feasibility.
