苦參根部化學成分分析

Phytochemical Studies on the Roots of Sophora flavescens

摘要


中藥材苦參,為豆科 (Leguminosae) 植物苦參的根。味苦,性寒,有清熱燥濕、袪風殺蟲之功用。根據國內外學者對苦參化學成分的研究發現,在其根中含有生物鹼及黃酮類成分。本實驗以採集自花蓮之苦參根部的甲醇抽出物,以各種色層分析方法 (矽膠管柱層析、Sephadex LH-20及HPLC) 進行分離純化,得到四個新化合物,5,7,2'4'-tetrahydroxy-5'-methoxy-8-lavandulylflavanone (11)、8-lavandulylkaempferol (18)、2-(2',4'-dihydroxy-5'-prenylphenyl)-5,6- methylenedioxybenzofuran (21) 及 2,4,2’,4’-tetrahydroxy-3-lavandulyl- 6-methoxybenzil (22),和二十五個已知化合物,包含二個 pterocarpans (1, 2)、二個 chromones (3, 4)、一個triterpene (5)、一個benzoate (6)、九個 flavanones (7~10, 12~16)、二個 flavonols (17, 19)、一個 2-aryl benzofuran (20)、一個medicagol (23)、四個 isoflavones (24~27)、一個coumarin (28) 和一個chalcone (29) 類化合物。這些化合物是經由質譜及核磁共振光譜的分析而確認。

The occurrence of alkaloids and flavonoids were reported previously in Sophora flavescens. We reinvestigated the constituents of S. flavescens which was collected from Hualien, Taiwan. A combination of several chromatographic techniques (silica gel, Sephadex LH-20 and HPLC) led to the isolation of four new compounds (including 5,7,2'4'-tetrahydroxy -5'-methoxy-8-lavandulylflavanone 11, 8-lavandulyl-kaempferol 18, 2-(2',4'-dihydroxy-5'-prenylphenyl)-5,6-methylenedioxybenzofuran 21 and 2,4,2’,4’-tetrahydroxy-3-lavandulyl-6-methoxybenzil 22) and twenty- five known compounds (including two pterocarpans, two chromones, one triterpene, one benzoate, nine flavanones, two flavonols, one 2-aryl benzofuran, four isoflavones one coumarin and one chalcone) from the MeOH extract of the roots of S. flavescens. Their structures were elucidated using mass and NMR spectral methods.
