斑蝥(mylabirs )其衍生物Norcantharidin與Dehydronorcantharidin對肝癌細胞株作用的探討

摘要


去甲斑蝥素( Norcantharidin,NCTD)是人工合成的一種新型抗癌藥物,為中藥斑蝥( Mylabris )體內抗癌有效成份斑蝥素的衍化物。在臨床應用上,已有去甲斑蝥素治療原發性肝癌的報導。但去甲斑蝥素對癌細胞抑制作用的詳細機轉尚未知,同時斑蝥的另一衍生物去水去甲斑蝥素( Dehydronorcantharidin,DNCTD )對肝癌細胞株的影響經由實驗,其亦會造成肝癌細胞株的死亡,只在於所需藥物濃度的不同。DNCTD在HA22T肝癌細胞株中的需10倍的NCTD劑量,在Huh7肝癌細胞株中的需5 倍的NCTD 劑量。時間的因素方面, NCTD 在24小時即引發細胞凋亡的作用,DNCTD 則需72 小時才有作用。至於其造成的影響,經由本實驗,由細胞之變化與Flow cytometry 分析等,巳可証實其作用為Apoptosis。至於其真正機轉尚需作進一步的分析探討。

Norcantharidin (NCTD) is a demethylated form of cantharidin with antitu-morproperties. It was used as a routine anticancer drug against hepatoma. How-ever,there is limited study on the effect of NCTD on human cancer cells, caused progressed to apoptosis within 48 hours in two human hepatoma cell lines:HA22T and Huh - 7. The precise mechanism of NCTD is still unknown. Dehy-dronorcantharidin(DNCTD), is another demethylated form of cantharidin with antitumor properties that caused progressed to apoptosis within 72 hours. DNCTD need 10 times dose than NCTD induced apoptosis in HA22T, and need about 5 times does in Huh7 cell lines. However the mechanism of anticancer is unknown,still need further investigation.
