嶺南野菊精油抑制骨肉瘤細胞遷移及貼附能力之分析

Analysis of Vernonia patula Crol in migration and adhesion of osteosarcoma cells

嶺南野菊,學名為Vernonia patula(Dryand.) Merr.,菊科(Asteraceae),斑鳩菊屬(Vernonia),屬一年生草本植物,原產於亞洲南部各地,株高約三十至七十五公分,臺灣藥用植物資源名錄記載其藥用成分可清熱利濕、散瘀消腫、解毒止瀉,可治感冒、頭痛、腹瀉、肝病、急性腸胃炎。本實驗室前期文獻已經證明嶺南野菊水萃精油(簡稱Crol)在抑制骨肉瘤細胞株143B和MG-63實驗中,具有劑量依賴性效果,實驗結果顯示,加入水萃精油24小時後,其半致死劑量分別為103.9±7.8 μg/mL(143B)、7.3±5.0 μg/mL(MG-63),且是透過G2/M arrest的方式來誘導癌細胞進行凋亡。本實驗將進一步研究嶺南野菊Crol對於腫瘤轉移的抑制效果,透過癌細胞遷移與貼附能力測試實驗,我們發現在50 μg/mL的作用濃度下,MG-63的遷移能力抑制率為16.6%(3 h)和28.3%(6 h),且是經由抑制基質金屬蛋白酶9(Matrix metallopoateinase, MMP-9)蛋白酶的活性,來抑制人類骨肉瘤癌細胞腫瘤遷移能力。以不同濃度(0、25及50 μg/mL),加入143B及MG-63的細胞培養液,作用4小時後結果發現,隨著精油濃度增加對細胞的貼附能力情形有降低的趨勢,在50μg/mL抑制率可分別達56.6%和34.8%。以上結果顯示嶺南野菊精油確實對人類骨肉瘤癌細胞,除了具有抑制細胞生長的作用外,也可抑制骨肉瘤細胞的轉移能力,值得進一步深入研究,以開發其臨床應用上的潛力。

Vernonia patula (Dryand.) Merr., is an annual herb native to southern Asia, with a height of about 30 to 75 cm. Taiwan's medicinal plant resources list records its medicinal ingredients for antipyretic and dampness, dispersing sputum and swelling, detoxification and antidiarrheal, and treating cold, headache, diarrhea, liver disease and acute gastroenteritis. The preliminary literature of our laboratory has proved that Vernonia patula water extract essential oil (abbrev. Crol) has a dose-dependent effect in inhibiting osteosarcoma cell line 143B and MG-63. The experimental results show that the lethal concentration 50% (LC_(50)) is 103.9±7.8 μg/mL for 143B cells, 57.3±5.0 μg/mL for MG-63 cells after 24 hours of adding Vernonia patula Crol, and the apoptosis of cancer cells was induced by G2/M arrest. This experiment will further study the inhibitory effect of Vernonia patula Crol on tumor metastasis. Through the test of cancer cell migration (wound healing assay) and adhesion ability, we found that the migration inhibition rate of MG-63 was 16.6 % (3 h) and 28.3% (6 h) at the concentration of 50 μg/mL, and the mechanism of inhibition may through inhibiting the activity of MMP-9 protease. At different concentrations (0, 25 and 50 μg/mL), 143B and MG-63 cells were added. After 4 hours of action, it was found that the adhesion ability of the cells decreased with the increase of Crol concentration, and the inhibition rate in 50 μg/mL was respectively up to 56.6% and 34.8%. The results above show that Vernonia patula Crol can inhibit the metastasis ability of osteosarcoma cells in addition to the inhibition of cell growth in human osteosarcoma cancer cells. It is worth further research to develop its potential for clinical applications.
