分析食用石蓮花水萃物對肝癌細胞株之毒殺作用

Cytotoxicity of water extract of Graptopetalum paraguayense in hepatocellular carcinoma cell lines

摘要


在無法以手術治療之晚期肝細胞癌的處理上,還須積極開發更多藥物治療的可能性。過去的研究結果指出,石蓮花萃取物具有抗氧化以及癌細胞毒殺活性。本研究使用超音波萃取方式製備石蓮花葉片之水萃物,使用沒食子酸以及芸香甘分別作為測量水萃物之總多酚與總類黃酮含量之標準品,並進一步探討石蓮花水萃物對不同的肝癌細胞株以及正常肝細胞之存活狀況的影響。實驗結果顯示,石蓮花水萃物總多酚濃度為215.28 GAE μg/ml,總類黃酮濃度為411.69 RE μg/ml。石蓮花水萃物對肝癌細胞株HepG2具有毒殺效果,水萃物處理48小時對HepG2存活的半抑制濃度IC_(50)為445.98 μg/ml;而對另一種肝癌細胞株Huh7以及大鼠正常肝細胞Clone 9而言,石蓮花水萃物處理48小時在濃度800 μg/ml 時尚未達到IC_(50)。然而石蓮花水萃物在低濃度的100與200 μg/ml範圍對於HepG2與Huh7細胞有明顯毒殺效果,但對於Clone 9不具有毒殺效果,顯示石蓮花水萃物能在對正常細胞沒有毒性的濃度之下對於肝癌細胞具有毒殺活性。本研究結果提供評估石蓮花水萃物作為臨床抗肝癌藥物可行性之參考。

In advanced stage hepatocellular carcinoma which is untreatable with surgery, more therapeutic drug candidates are to be explored. Previous reports had shown that the extract of Graptopetalum paraguayense has antioxidative and antiproliferative activities. In this study, the water extract of Graptopetalum paraguayense (WEGP) was prepared by ultrasonic extraction, and the total polyphenols and total flavonoids content in the extract were measured using gallic acid and rutin as standards, respectively. Cytotoxic effect of WEGP was assessed in two human hepatocellular carcinoma cell lines HepG2 and Huh7 and rat normal liver cells Clone 9. The total polyphenols content in WEGP is 215.28 GAE μg/ml, and the total flavonoids content is 411.69 RE μg/ml. WEGP has cytotoxic activity against HepG2 with an IC_(50) of 445.98 μg/ml after 48 h treatment, but did not reach its IC_(50) at 800 μg/ml for Huh7 and Clone 9 cells. However, in low concentration range of 100 and 200 μg/ml, WEGP has significant cytotoxic activity against HepG2 and Huh7, but not for Clone 9 cells. We have demonstrated a selective cytotoxic activity of WEGP against hepatocellular carcinoma cell lines but not against normal liver cells under certain concentration range. Our results provide useful information of WEGP for possible application as a therapeutic drug against hepatocellular carcinoma.
