Applying DNA sequences on the mitochondrial genome are frequently undertaken for population assignments and inferences of the geographic area. Although it is common to assign the individual to a broad continental population for exclusory purpose, a finer resolution to subpopulations can provide much more information. The webtool of the PopInference was launched as the user-friendly tool to assign an individual into the most supported population and subpopulation. PopInference was built based on the k-nearest-neighbor (KNN) and k-weighted-nearest-neighbor (KWNN). Algorithms and using the genetic distance of the control region of the mitochondrial DNA (mtDNA) as the distance matrix. In this study, six cases from the case study were collected from the Institute of Forensic Medicine, Ministry of Justice. The cases in the case study were applied to webtool of population assignment to test the performance and the availability of the PopInference. Additional 399 mtDNA sequences were included as the extended sample set, including the British, Han, Indonesian, Vietnamese and Thai subpopulations. Increased samples of the British and Han subpopulations were used to test the accuracy result of the subpopulation assignment when the sample number of the subpopulation raised. The accuracy results of ethnic inferences for the British and Han subpopulations were higher in the KNN and KWNN algorithms when increasing the sample number of the both two subpopulations. Subpopulations of the Indonesian, Vietnamese and Thai were applied to complete the data of the subpopulation assignment for further application in Taiwan.
