γ-射線照射蔓綠絨組培苗莖段之誘變研究

Induction of Morphological Variants from Stem Node Explants of Philodendron after Treatment with γ-rays

摘要


利用組織培養配合γ-射線照射的處理方式,誘導蔓綠絨(Philodendron)產生變異,期由變異體中發現具有優良性狀的個體以做為品種開發的基礎。結果顯示,泡泡蔓綠絨莖段培養於含有么2.0mg/l BA (N^6-benzyladenine)或0.5mg/l TDZ(thidiazuron)的MS液態培養基預處理2星期,再接種於含有1.0mg/l BA及0.2mg/1 NAA (α-naphthaleneacetic acid)的MS固態培養基培養6星期後,每個培植體可誘導萌發5.8與5.6芽體,效果較佳。泡泡(P cv. 'Tuxla')、綠帝王(P. cv. 'Imperial Green')、鋸齒(P. cv. 'Pluto')及紅公主(P. cv. 'Red Princess')蔓綠絨莖段培養於1.0mg/l BA及0.2mg/l NAA的MS固態培養基,均可達大量繁殖的目的。紅公主蔓綠絨經γ-射線照射處理後產生葉片呈黃色、淡黃色、白綠斑紋或黃綠斑紋的變異植株;這些黃色、淡黃色或黃綠斑紋的異常植株,分離培養時均可大量繁殖,而白綠斑紋的異常植株,分離培養後往往消失此性狀。泡泡、綠帝王與鋸齒蔓綠絨經γ-射線照射處理後,並無顯示可見的形態變異。

Stem node explants derived from in vitro propagated shoots of four cultivars of Philodendron viz. 'Tuxla', 'Imperial Green', 'Red Princess', and 'Pluto' were irradiated with γ-rays (20 and 40 Gy) to induce variation. The explants were pre-cultured in liquid Murashige and Skoog's (MS) medium supplemented with 2.0 mg/l N^6-benzyladenine (BA) or 0.5 mg/l thidiazuron (TDZ) before irradiation. After irradiation, the explants were cultured on MS medium supplemented with 1.0 mg/l BA + 0.2 mg/l (-naphthaleneacetic acid (NAA) for production of multiple shoots. Morphological variants were observed only in cv. 'Red Princess'. Other three cultivars, though could be successfully propagated in vitro, failed to show any morphological variation after treatment with γ-rays.
