The fine mapping and cloning quantitative trait loci (QTLs) responsible for traits of agronomic importance in rice is known as the most general stategy in plant genomics and provides a foundation to select new rice varieties whith high yield quality, resistance via marker-assisted selection (MAS) in rice breeding programs. Chromosome segment substitution lines (CSSLs) are one of the most powerful tools for the detection and precise mapping of QTLs. In this paper, the principle of developing the CSSLs was described, the research progress of consturction and application was reviewed, and the prospect was discussed.
