用含有轉座子Tn5的銅綠假單胞杆菌PAO1826(pMO75::Tn5,Km(上標 r))誘變我國馬鈴薯青枯菌小種3號菌株PO41,獲得胞外多糖正常産生的接合子6000多個,經SDS-PAGE電泳篩選出胞外蛋白減少1~9種的菌株21株。經測試其中20株爲原養型菌株,1株爲營養缺陷型菌株。這21株突變株在煙草葉片上呈現典型的過敏性反應。用莖部毛細管滴注法在馬鈴薯植株上進行致病力測試的結果表明,胞外蛋白發生減少的21株突變株中有20株致病力均不同程度下降,通過分析突變株胞外蛋白缺失種類與致病力強弱的關係,初步確定59.5ku、55ku和47ku的胞外蛋白在青枯菌致病過程中起著十分重要的作用。

English Abstract

A wild type strain (P041) of race 3 of Ralstonia solanacearum was mated with Pseudomonas aeruginosa strain PA01826 (pM075:: Tn5, Km(superscript r)), more than 6000 transconjugants were transferred to TZC+Km medium to recognize production of EPS. Culture supernatants of irregular round, white or pink, fluidal colonies were analyzed by SDS-PAGE, and 21 mutants deficient of 1 to 9 protein(s) obtained. Twenty mutants were tested to be prototrophic type strains, all of the 21 strains could cause hypersensitive reaction on tobacco leaves. Virulence test by stem-puncture inoculation revealed that the 20 mutants were less virulent on potato plant to different extent. Comparison of virulence of these mutants suggested that 3 kinds of proteins (59. 5ku, 55ku, 47ku) played key role in Ralstonia solanacearum pathogenesis.
