Chinese Abstract

目的:探讨高浓度表皮生长因子(EGF)对小鼠肝细胞信号转导的动力学改变。方法:从细胞整体水平的角度出发,采用双向电泳及放射性同位素标记技术,并结合相关数据库资源和双向电泳专用分析软件,分析小鼠肝细胞在高浓度的EGF信号持续刺激不同时间得到的EGF信号蛋白在EGF信号转导过程中其磷酸化水平发生的动力学变化。结果:在高浓度EGF持续刺激下,小鼠肝细胞内的某些结点信号蛋白如Eps15和ERK2的磷酸化动力学曲线表现为持续活化状态。结论:在高浓度的EGF持续刺激下,EGF信号网络系统中的某些信号蛋白将会发生一定的改变,这些改变可能与肿瘤的发生、发展有密切关系。

English Abstract

Objective: To study the change of dynamics of high concentration EGF signalling transduction system in mice hepatocytes. Methods: 2-DE method, PDQuest 2D, Swiss-Prot/TrEMBL and PPDB etc. were used to assay the phosphorylated proteins related to high concentration EGF signaling transduction in the whole hepatocytes. Dynamic curves of relative intensity versus time were drawn after analyzing changing of phosphorylating level versus varying time of phosphorylated proteins. Results: Some node-proteins of EGF such as Eps15 and ERK2 showed constant activation under the constant stimulation of high concentration EGF. Conclusion: Under the constant stimulation of high concentration EGF, the activity of some signal proteins in EGF signalling transduction system will change, which is probably related to the development of tumor.
