目的 研究银杏叶提取物对人乳腺癌MCF7细胞脂肪酸合酶(fatty acid synthase, FAS)的抑制作用。方法 MTT法测定人乳腺癌MCF7细胞增殖速度,采用超速离心技术部分纯化人乳腺癌MCF7细胞FAS。不同浓度银杏叶提取物与FAS相互作用不同时间后,加入底物,用分光光度法观察银杏叶提取物对FAS的抑制作用。结果 银杏叶提取物对人乳腺癌MCF7细胞的增殖有一定的抑制作用。人乳腺癌MCF7细胞FAS被部分纯化,三种浓度银杏叶提取物(0.1,0.5,1g/L)与FAS在催化前相互作用0、5和10min,对FAS活性的抑制率分别为14.60%、20.30%和32.75%(0.1g/L);33.50%、40.30%和84.03%(0.5g/L);90.60%、88.30%和88.05%(1g/L)。结论 银杏叶提取物对人乳腺癌MCF7细胞FAS具有抑制作用;作用强度既依赖于抑制剂浓度,又依赖于抑制剂与酶相互作用时间。

English Abstract

Objective: To study the inhibition to Fatty Acid Synthase from human breast cancer cell with extract of ginkgo leaf (GL) in Vitro. Methods: The human breast cancer MCF7 cell proliferation was determined by MTT spectrophotometry, the human breast cancer MCF7 cell FAS was partially purified by ultra-centrifugation. The FAS was interacted with different concentrations of GL extract with different times before catalyzing. Then the substrates of FAS were added to the reaction system. The inhibitory activities of GL extract against the FAS were investigated using spectrophotometric assays. Results: GL extract inhibited proliferation of human breast cancer MCF7 cell. The human breast cancer MCF7 cell FAS was partially purified. The inhibitory activities of GL extract (0.lg/L) at 0、5、and 10 minute on FAS were l4.60%、20.30%、and 32.75%, respectively. The inhibitory activities of GL extract (0.5g/L)) at 0、5、and 10 minute on FAS were 33.50%、40.30% and 84.03%, respectively. The inhibitory activities of GL extract (1g/L) at 0、5、and 10 minute on FAS were 90.60%、88.30% and 88.05%, respectively. Conclusion: GL extract inhibits the FAS from human breast cancer MCF7 cells. The inhibitory activities depended on the concentrations of GL extract and the interaction times between FGL extract and FAS before catalyzing.
