目的 研究血管内皮生长因子(VEGF) B mRNA在乳腺癌组织中的表达及意义。方法 采用逆转录-聚合酶链反应技术对手术的47例乳腺癌标本和11例乳腺良性病变标本中VEGF-B mRNA的表达进行检测。结果 在良恶性乳腺组织中均检测到VEGF-B167、186 mRNA表达,VEGF-B167在乳腺癌和良性乳腺组织中100%表达,VEGF-B186在乳腺癌和良性乳腺组织中的表达率分别为78.7%和54.5%。VEGF-B167在乳腺癌和良性乳腺组织中表达水平分别为(0.222±0.078)和(0.176±0.062),两者差异无显著性(P=0.077)。VEGF-B186在乳腺癌和良性乳腺组织中表达水平分别为0.167(0.080~0.219)和0.071(0.000~0.156)。乳腺癌中表达水平明显升高(P=0.010)。VEGF-B167、186 mRNA表达与乳腺癌患者年龄、肿块大小、淋巴结转移状况、肿瘤分期及雌、孕激素受体状况之间无明显相关性。结论 乳腺癌组织中VEGF-B186 mRNA袁达水平上调,而VEGF-B167 mRNA表达水平无明显上调,提示VEGF-B186可能在乳腺癌中起着某些生物学作用。

English Abstract

[Objective] To study the expression of vascular endothelial cell growth factor (VEGF)-B mRNA in breast cancer tissues and its correlation with clinicopathological factors. [Methods] The expression of VEGF-B mRNA were analyzed by reverse transcription-polymerase chain reaction (RT-PCR) in the specimens from 47 patients with breast cancer and 11 patients with benign breast disease. [Results] VEGF-B167, 186 mRNA were detected in both malignant and benign breast tissues. VEGF-B167 expressed in all malignant and benign breast tissues, while VEGF-B186 expressed in 78.7% breast cancer tissues and 54.5% benign breast tissues. The expression level of VEGF-B167 was (0.222±0.078) and (0.176±0.062) in breast cancer tissues and benign breast tissues, respectively, with no significant difference (P=0.077). While the expression level of VEGF-B186 was higher in breast cancer tissues, 0.167 (0.080~0.219), than in benign breast tissues, 0.071 (0.000~0.156) (P=0.010). Moreover, we observed there was no correlation between the expression of VEGF-B167, 186 mRNA in breast cancer tissues and patients' age, tumor size, lymph node metastasis, tumor stages, estrogen or progesterone receptor status. [Conclusion] The expression of VEGF-B186 mRNA was up-regulated in breast cancer tissues, while the expression of VEGF-B167 mRNA was not, thus which suggested VEGF-B186 may have some biologic activity in breast cancer.
