Chinese Abstract

Objective: To investigate the changes of TGF-β1 mRNA in adventitial cells and the effects of Oleum Curcumae Aromaticae oil injection on injuried rat arteriesafter balloon angioplasty. Methods: The left common carottid artery in rats was denuded with a balloon catheter. Cross-sections from the injuried segments of the injuried rat arteries at 7 days were processed for the expression of TGF-β1 mRNA in adventitial cells study by in situ hybridization technology and computer-assisted picture andlysis system. Histological and morphological study were processed by computer-assisted picture analysis system at 14 days. Results: Seven days after balloon injury, Arteries of sham-opration group displayed low-level expression of TGF-β1 mRNA in Adventitial cells. Compared with sham-opration group, adventitial cells of model group showed dramatic upregulation of TGF-β1 mRNA that the positive is perinuclear buffy reaction. Oleum Curcumae Aromaticae oil injection caused a marked reduction of expression of TGF-j3l mRNA in adventitial cells.14 days after balloon injury, Lumen area, EEL perimeter, IEL perimeter of model group were decreased significantly (P<0.01) than those of sham-operation group. Compared with model group, maximal intimal thickness, neointima area of Oleum Curcumae Aromaticae oil group were decreased significantly (P<0.01), but with no differences compared with those of the shamoperation group (P>0.05). Lumen area, EEL perimeter, IEL perimeter increased markedly (P<0.01) up to that of the shamoperation group (P>0.05). Conclusion: The expression of TGF-β1 mRNA in adventitial cells is a major cause of restenosis after arterial injury. We concluded that Oleum Curcumae Aromaticae oil injection inhibits neointimal formation and vascular remodeling after arterial injury by inhibiting expression of TGF-β1 mRNA in adventitial cells.
