Chinese Abstract

Objective: To observe the change of tumor necrosis factor-α by preconditioning with Panax notoginseng saponins (PNS) on myocardial ischemia- reperfusion (IR) injury in rats. Methods: The model was established by left anterior descending coronary artery ligation under thoracotomy, preconditioning immediately preceded 45 min after left coronary artery occlusion followed by 120 minute's reperfusion. Twenty-five male Sprague Dawley rats were divided into four groups: sham operation group (n=4), IR group (n=7), IPC group (n=7) and preconditioning with PNS group (n=7). The levels of creatine phosphokinase-MB (CK-MB1 and tumor necrosis factor-α (TNF-α) in serum were measured. Results: The levels of serum CK-MB in the IPC group and preconditioning with PNS group were significantly lower than that in IR group (2304.29±540.04 U/L and 5098.43±1268.18 U/L vs. 17925.71±2022.74 U/L, P<0.05, respectively). The levels of serum TNF a in the JPC group and preconditioning with PNS group were significantly lower than that in IR group (1.08±0.08 ng/ml and 1.11±0.18 ng/ml vs. 1.51±0.24 ng/ml, P<0.05, respectively). Conclusion: Preconditioning with Panax notoginseng saponins can attenuate myocardial ischemia-reperfusion injury through restraining the levels of serum TNF-a, but the protection effect is inferior to IPC.
