諾麗果發酵產物對BALB/c小鼠脾臟細胞Th1/Th2細胞激素分泌之影響

Immunomodulatory Effects of the Fermented Fruit of Noni, Morinda citrifolia, on the Secretion of Th1/Th2 Cytokines by Splenocytes from BALB/c Mice

摘要


諾麗(Morinda citrifolia L., noni)是一種藥用植物,常被宣稱對某些疾病具有療效。本研究利用體外實驗模式,探討台灣產諾麗果(noni fruit)對BALB/c小鼠脾臟細胞分泌輔助T-細胞類型1(helper T-cell type 1, Th1)及2(Th2)細胞激素分泌的影響。實驗以不同發酵程度之諾麗果製品,與初代脾臟細胞共同培養,評估這些樣品對脾臟細胞增生的影響,並檢測脾臟細胞分泌Th1/Th2細胞激素的變化。實驗結果顯示,相較於未發酵或發酵30天的諾麗果樣品,發酵60及90天的樣品在低濃度時(≦500 μg/mL)對小鼠脾臟細胞具有促進增生的作用;從細胞激素分泌的結果顯示,諾麗果樣品會促使小鼠脾臟細胞分泌較高比例Th2類型的細胞激素,尤其以發酵90天的樣品最為顯著。綜合本實驗的結果,推測諾麗果樣品對小鼠脾臟細胞的增生和細胞激素的分泌具有調節潛力。

Morinda citrifolia L. (noni) is a plant used in traditional medicine. Many therapeutic claims have been made for noni products. To test the immunomodulatory effects of noni fruit on helper T-cell type 1 (Th1 ) and Th2 cytokine secretions, we prepared different fruit products of M. citrifolia cultivated in Taiwan and applied them to primary splenocyte cultures from female BALB/c mice. The noni fruit products were first prepared by fermenting the ripe fruit for 0, 30, 60, and 90 days, respectively. The results showed that noni fruit products which were naturally fermented for 60 and 90 days produced increased proliferation of splenocytes at the indicated concentrations (≦500 μg/mL) compared to products from the control and 30-day fermentation. Further analysis of Th1/Th2 cytokines produced by splenocytes showed that noni products, particularly the 90-day fermented product, increased Th2 cytokine secretion. This study suggests that fermented noni fruit products exhibit an immunomodulatory potential via increasing the proliferation and secretion of Th2 cytokines by primary splenocytes from BALB/c mice.
