Effects of Angiotensin Ⅱ on the Proliferation and Fibronectin Production of Glomerular Mesangial Cells

血管收縮素Ⅱ對腎絲球環間質細胞之增生及纖維蛋白元製造之影響

摘要


血管收縮素Ⅱ(以下簡稱Ang-Ⅱ)是一種極強的血管收縮劑,在腎絲球硬化過程中扮演極重要之角色。由於腎絲球硬化的特徵是腎絲球細胞及細胞外間質的異常,因此本研究主要探討Ang-Ⅱ對腎絲球環間質細胞之增生及製造纖維蛋白元(一種主要的細胞外間質)的影響。研究方法中纖維蛋白元mRNA是以西方點墨法測定,而纖維蛋白元是以北方點墨法測量。研究結果(1)Ang-Ⅱ可以增加細胞[H^3]-thymidine之攝取量,且其攝取量隨Ang-Ⅱ使用劑量增加而增加。(2)Ang-Ⅱ可以增加細胞的數量,且其數量隨Ang-Ⅱ刺激時間之增長而增加。(3)Ang-Ⅱ可以增加細胞纖維蛋白元mRNA之表現量及其蛋白質之產量,且其產量隨Ang-Ⅱ刺激時間之增長而增加。以上結果顯示Ang-Ⅱ對於腎絲球環間質細胞有強力的刺激作用,可以增加其細胞的增生及纖維蛋白元之產生。

Angiotensin Ⅱ (Ang-Ⅱ) is a potent vasoactive hormone which plays an important role in the pathogenesis of glomereulosclerosis. Since glomerulosclerosis is characterized by alterations in both cellularities and the extracellular matrix, we investigate the effects of Ang-Ⅱ on the cellular proliferation and fibronectin production of glomerular mesangial cells. Fibronectin was measured by Western blot analysis and fibronectin mRNA was measured by Northern blot analysis. The results were as follows: (1) Ang-Ⅱ increased [3H]-thymidine uptake of mesangial cells in a dose-dependent manner. (2) Ang-Ⅱ increased the number of mesangial cells in a time-dependent manner. (3) Ang-Ⅱ enhanced fibronectin mRNA expression and its production of mesangial cells in a time-dependent manner. Our results indicate that Ang-Ⅱ is an active stimulant for the proliferation and fibronectin production of glomerular mesangial cells.
