摘要


飛行人員自行服用降血壓藥Propranolol、利尿劑Furosemide及降血糖藥Glibenclamide等有其潛在的危險性,一來可能當時的身體狀況不適合飛行,而且藥物本身也可能干擾生理機能,降低操作能力,影響飛安甚鉅,故這些藥物的分析是航空毒物學檢測中的重要項目之一。本研究利用液相層析法來測定血清及尿液中之Propranolol、Furosemide及Glibenclamide,檢體經鹼化或酸化後,以乙酸乙酯抽取,抽取液注入高效能液相層析儀,經由移動相-氰甲烷/磷酸鹽緩衝液(pH3.2,含有0.05%壬胺)的帶動,通過微徑的C18逆相分離管,再由螢光檢測器偵測。本法準確、快速,靈敏度又高,未來可廣泛應用於航空毒物學上具有螢光性質藥物之鑑定分析。

Self-medication of propranolol, furosemide or glibenclamide has two potential dangers to the flyer. First, the conditions being treated will likely preclude safe flying, and second, the adverse effects of the drugs themselves may affect flight activities. This paper presents a method for the simulataneous analysis of propranolol, furosemide and glibenclamide in serum and urine. The specimens, alkalinized with saturated sodium borate or acidified with hydrochloric acid, are extracted with ethyl acetate. The extracts are eluted from a reversed-phase microbore column with a mobile phase consisting acetonitrile/phosphate buffer (pH3.2, containing 0.05% nonylamine), using a gradient system. The eluted drugs are detected by a fluorescence detector. The recoveries and within run and day to day precision of the method have been investigated and found to be good. The method is fast, simple, and sensitive and will be applied in aviation toxicology for the determination of drugs with fluorescence properties in biological samples.
