不同去乙醯化度幾丁聚醣包覆檸檬精油微膠囊之製備及其緩釋性之探討

Preparation and Sustained Release Properties of Various Deacetylated Chitosan Microcapsules Containing Volatile Essential Lemon Oil

摘要


本研究以蝦殼為原料,經酸鹼處理後製成幾丁質,再經高溫熱鹼處理製備不同去乙醯化度幾丁聚醣作為微膠囊之殼材。將這些幾丁聚醣溶於醋酸水溶液中形成幾丁聚醣膠體,再以液中硬化法製成包覆揮發性精油的膠囊,探討不同去乙醯化度幾丁聚醣之膠體黏度與形成膠囊的可能性以及微膠囊中精油的緩釋行為。結果顯示經滴定與黏度測試後幾丁聚醣膠體的去乙醯化度可達88%,即使再延長熱鹼處理時間也無法大幅提升去乙醯化度,但其膠體黏度卻有顯著下降之趨勢。使用去乙醯化度77%、82%、88%三種幾丁聚醣作為殼材包覆揮發性檸檬油,比較檸檬油包覆前後的制放情形,結果發現色覆後的精油制放呈現定時定量的釋出,且釋出時間相較於未經膠囊包覆之樣本延長一倍以上,達到良好的緩釋效果。之後選擇三者中緩釋效果最佳的88%去乙醯化度幾丁聚醣膠囊,探討使用硫酸、乙二醛、葡萄糖三種交鏈劑處理微膠囊後的精油制放率,發現三者均可得到更佳的緩釋效果。

In this study, chitosan microcapsules containing volatile essential oil were prepared with various deacetylated chitosan wall materials by Orifice Method. The relation of deacetylated degree and viscosity of chitosan; the formation conditions of chitosan microcapsules and sustained release effect of volatile essential oil were investigated. The results indicated that the viscosity of chitosan was decreased with increasing the deacetylated degree of chitosan. The chitosan microcapsule can be obtained at the conditions of deacetylated degree 45%-90% of chitosan. The sustained release effect of volatile essential oil was improved over 100% by microencapsulated with chitosan wall membrane.
