目的：筛选脱牛心包衍生膜材料的制备方法，观察其组织结构和力学性能，探讨作为支架材料的可能性。方法：分别采用0.1 mol/L NaCl+0.5% Triton X-100、0.1 mol/L NaCl+0.25% Trypsin、0.25% Trypsin+0.5 % Triton X-1 00、0.1 mol/L NaCl+ 0.25 % Trypsin + 0.5% Triton X-100对新鲜牛心包进行脱细胞处理，脱细胞牛心包经过碳化二亚胺(EDAC)交联。观察脱细胞牛心包基质材料的组织结构、测定其理化性质。结果：经 0.25% Trypsin+ 0.5 % Triton X-100酶联脱细胞处理获得的脱细胞牛心包内无细胞成分，其双层结构及理化性能保存良好。结论：0.25% Trypsin+0.5% Triton X-100酶联脱细胞法能有效地脱去牛心包内的细胞成分，获得的脱细胞牛心包的组织结构和理化性能符合生物支架材料的要求。
Objective: To choose the best procedure on preparation of acellular bovine pericardium (BP) bio-derived material and to validate the possibility of its use as brace material. Methods: The bovine pericardiums were treated with 0. 1mol/LNaCI+0.5%TritonX-100(group Ⅰ),0.1mol/LNaCl+0.25%Trspsin(group Ⅱ),0.25%Trypsin + 0.5%Triton X-100(group Ⅲ), 0.1mol/LNaCl + 0.25% Trypsin+0.5% Triton X-100 (group Ⅳ) respectively. The histological structure and mechanics of modified BP were compared between tile four groups. The hest BP in the groups was treated with EDAC, which then compared the surface structure with the collagen membrane and the Bin-Gide membrane. Results: Histological examination showed cellular constituents were removed in all groups. The mechanics(the shrinkage temperature, the tensile strength, the maximal strain and the elongation properties) of group Ⅲ were better than others (P＜0.05). The mechanics of group Ⅲ were improved alter treated with EDAC. Electron microscopy showed that the BP treated with EDAC hail two different surfaces, which was similar to the Rio-Gide membrane. Conclusions: 0.25% Trypsin + 0.5% Triton X-100 can extract all cellular constituents and the acellular bovine pericardium bin-derived material satisfied the aced for brace.
