Chinese Abstract

Objective: To study the expression and significance of p65, an important subtype of nuclear transcription factor KB (NF-κB) and its upstream kinase IKKα in malignant transformation of hamster boccal mucosa. Methods: We established the animal model of malignant transformation in hamster buccal mucosa induced by DMBA (0.5%). Twelve paired specimens including normal buccal mucosa, epithelial hyperplasia, epithelial dysplasia and squamous cell carcinoma (SCC) were subjected to western blot for anal\sis p65. The expression of IKKα was ohser4ed in the four different groups by immunohistochemical evaluation. Results; In normal buccal epithelia and hyperplasia epithelia, evident Western blotting hand of p65 was not fonnd, and there was no significant difference between them(P>0.05); the expression of IKKα in normal buccal pooch mucous membrane was almost negative, and evident plasm staining was not found in the cellular layers of mucous membrane epithelia. The staining of hyperplasia epithelia was similar to that of normal buccal epithelia. With the occurrence of epithelia dysplasia, the expression of p65 gradually increased, and significant difference could he found when comparing with normal buccal epithelia and hperplasia epithelia(P<0.01); at the same time, comparing with normal buccal mucosa and epithelial hyperplasia, the positie staining intensit of IKKα raised with the increase of positie cells, which had some relations with the degree of epithelia dysplasia. In SCC, p65 was expressed to a higher level comparing with normal buccal mucosa and dysplasia(P<0.01), dole the positive expression of IKKα increased significantly with the broad positive expression. Significant differences could he found when comparing with normal boccal epithelia and epithelial dysplasia (P<0.01). Conclusions; NF-κB p65 is strongly activated in malignant transformation of hamster buccal mucosa. P65 and IKKα expressed abnormally in malignant transformation of hamster buccal mucosa.
