Prior Induction of Heme Oxgenase-1 with Cordycepin Attenuates Renal Ischemia and Reperfusion Injury in Rats

已有學者指出冬虫夏草可保護腎臟免於aminoglycoside導致的傷害,可增加紅血球細胞膜上SOD的活性並降低MDA量。許多腎臟疾病包括glycerol引發急性腎衰竭,aminoglycoside引發急性腎衰竭,cisplatin引發急性腎衰竭,及缺血再灌流導致之腎衰竭被發現可誘發heme oxygenase-1表現增加。在CYP代謝之一連串反應中可產生大量氧化自由基。這些氧化自由基會導致之腎臟傷害。本實驗證實冬虫夏草前投予四週[e0.8gm/day(0.2gm/ml)],可經由調控老鼠腎臟heme oxygenase-1蛋白表現增加來降低因缺血再灌流導致之腎臟傷害,而非經由CYP代謝減少所產生。

Background and Purpoes: Some investigators have pointed out that cordycepin could protect the kidneys from aminoglycoside-induced injury, increase superoxide dismutase (SOD) activity on red blood cell (RBC) membranes and reduce plasma malondialdehyde (MDA) levels. Many renal diseases, including glycerol-induced acute renal failure (ARF), aminoglycoside-induced ARF, cisplatin-induced ARF and ischemic reperfusion ARF, were found to be involved with catalytic iron release, and upregulation of the heme oxygenase-1 gene. Cytochrome P450 (CYP), a family of heme monooxgenases, catalyze the oxidation of a wide range of endogenous and exogenous compounds. Superoxide anion, hydrogen peroxide and hydroxyl radical can be formed in the CYP catalyzed reactions. These reactive oxygen metabolites (ROM) will cause pathophysiological damage to the kidneys. Methods: In order to clarify the protection effect of cordycepin on ischemic reperfusion (IR) renal injury resulting from upregulation of heme oxygenase-1 or decreased degradation of CYP, we fed cordycepin 0.8 g/day (0.2 g/ml) to rats, with right nephrectomies, for four weeks prior to IR injury. Results: We found that the renal function was better preserved in the cordycepin pretreated groups. The Western blotting technique showed upregulation of heme oxygenase-1 in cordycepin pretreated groups and the CYP level was not altered. Conclusions: These results provide direct evidence that prior induction of heme oxgenase-1 with cordycepin attenuates renal ischemia and reperfusion injury in rats.
