柳杉皮部之化學成分研究

Studies on the Chemical Constituents of the Bark of Cryptomeria japonica D. Don.

摘要


柳杉(Cryptomeria japonica (L. f.) D. Don.)屬於杉科,原產於中國大陸及日本,於日治時代引進台灣,現為全島海拔500~1800公尺間之主要造林樹種,是木造房屋之主要材源。本研究係將柳杉皮部,以甲醇浸泡得到之萃取物,運用矽膠薄層分析、矽膠管柱層析、高效率液相層析儀及再結晶等方法進行分離純化。單離之純化合物經測定紅外線光譜、氫核磁共振光譜、碳核磁共振光譜與質譜後,經推測若為已知化合物者,與文獻值比對確定其結構。若為新化合物,依需要進行二維核磁共振光譜實驗,配合化學反應等方法確定其結構。 本實驗鑑定得到共170個化合物,分別為: 一、單萜類(Monoterpenoid)化合物1個 二、倍半萜類(Sesquiterpenoid)化合物2個 三、雙萜類(Diterpenoid)化合物104個 四、雙萜雙聚類(Diterpenoid dimer)化合物12個 五、C35骨架類(C35 skeleton)化合物10個 六、木酚素類(Lignoid)化合物7個 七、類固醇類(Steroid)化合物10個 八、其他類(Others)化合物24個 共計有64個新化合物和2個從天然界首次分離得到之化合物。共計32種骨架,其中7種為新骨架。

Cryptomeria japonica (L. f.) D. Don., a member of the Taxodiaceae, is a protophyte in Mainland China and Japan. It was domesticated into Taiwan in 1906 and is distributed at altitudes of 500-1800 meters. It is a common building material. The methanol extract from the bark of C. japonica was concentrated to give a residue which was partitioned between ethyl acetate and H2O. The ethyl acetate layer was separated by column chromatography on silica gel, preparative thin layer chromatography, flash column chromatography, and high performance liquid chromatography (HPLC) to give one hundred and seventy compounds. The known compounds were identified by IR, NMR, and Mass spectral analysis and comparison with authentic samples or those reported data in literatures. The structures of new compounds were further elucidated by 2D NMR spectral and chemical methods. Among these compounds, there are sixty four new compounds and two isolated compounds for the first time from natural sources. All these compounds belong to thirty two skeletons, in which, there are seven novel skeletons.
