利用高效能液相層析法結合固相萃取技術來分析人參及靈芝中有效成分並建立化學指紋雷達圖

摘要


隨著近年來,中草藥應用已逐漸受到中西方在臨床醫療的重視,然而,中草藥的來源往往對療效品質有甚大的影響。因此,本實驗即選擇了自古以來已在中國藥材上佔有舉足輕重地位的人參與靈芝做為進行實驗分析的對象。實驗的目的主要在分析兩種不同產地的人參和靈芝種類,並比較其所含具療效的化學成分差異及含量,同時藉以建立化學指紋雷達圖譜,並可有效地達到品質控管的目的。 本研究分析方法採用固相萃取法(Sep-Pak C18 Cartridges),先以提取G-115人參膠囊中之人參皂苷做為人參樣品之標準品比對,而人參和靈芝樣品的化學成分萃取則分別以溶劑萃取(70%乙醇)與固相萃取( Sep-Pak C18 Cartridges)兩種方法進行萃取步驟。最後將所得到之萃取液再以高效能液相層析儀(HPLC)結合紫外光-可見光偵測器 (UV detector)進行分析,同時藉以比較其層析圖譜間的差異性。 有鑒於以往考慮樣品的回收率問題以及分離時間、效率等因素,本實驗採用固相萃取法配合分段收集的方式進行萃取。除了可比較各分段萃取液中化學成分組成外,亦可用於雷達圖的指紋圖譜建立。根據所選定之吸收峰須具備其為已鑑定之化學成分、分離效果較佳及訊號較明顯者特性,因此,依照萃取的樣品次序則採用三次分析結果進行分析統計。將所選定特殊波峰之面積將之加總後,即可製作成化學指紋雷達圖譜,因此,便可於日後作為判別這兩類藥材的種類與產地依據。

In recent years, the application in medical treatment of Chinese herbal medicine is respected gradually in medical science field. However, the sources of Chinese herbal medicine will affect the curative effect, quality and characters highly. Base on this study, we choose Ginseng and Ganoderma lucidum which play an important role in tradition Chinese herbal medicine. The target of this research is aimed at two kinds of Ginseng and Ganoderma lucidum which came from different location. By comparing the difference between the content and quality of the chemical components to be the main point, furthermore establishing radar fingerprint atlas for these Chinese herb medicine is a purpose to reach quality control demonstrable effect. In this research, extracting the Ginsenosides in the G-115 capsules as standards for Ginseng sample by using Sep-Pak C18 Cartridges. Otherwise, choosing two kinds of Ginseng and Ganoderma lucidum which come from different producing areas. Ginseng and Ganoderma lucidum of chemical components were extracted by solvent(70% ethanol) and solid phase extraction technique(Sep-Pak C18 Cartridges). Then the extracts were analyzed and separated by high performance liquid chromatograph that combined with ultraviolet-visible absorbance detector is followed the comparing the difference of chromatograms with each other. Because of recovery ratio, separation resolution and retention time, this reaserch use Sep-Pak C18 Cartridges extration methods and collect extractctions with fractional mode to reach effective analysis. Besides comparing the difference of chromatograms in each portion, using the analytical result to extablish radar fingerprint atlas successfully. First, selecting characteristic peaks is the main step. Second, comfirming chemical components, better separation resolution and stronger absorbance signals are referances factor. According to the characteristic peaks, compile to statistics by collecting three times result and sum of the integral areas of these signals in each portion. At last, all of statistics result could make radar fingerprint atlas which can offer the enough information which related to different Chinese herbal medicine sources and the place of origin.
