Chinese Abstract

過敏性氣喘屬於第一型過敏反應,患者體內免疫平衡傾向Th2反應,為一種免疫失衡的疾病。苦蕎麥在台灣屬於新興作物,在文獻中已發現其具有抗氧化、降血糖、降血脂及預防便秘等功效,但對於調節過敏性氣喘的功能則是尚未明瞭,因此本研究目的為探討攝食台灣苦蕎麥對免疫調節功能之影響,分別進行體外的細胞實驗和氣喘BALB/c小鼠動物實驗。分析苦蕎麥熟粉基本成分與類黃酮含量後進行體外實驗,結果發現蕎麥冷水萃取物在低濃度時具有抗發炎效果,進一步分析蕎麥中類黃酮成份與細胞激素分泌量的相關性顯示槲皮素為刺激細胞激素分泌的主要貢獻者,其中以苦蕎熟粉的槲皮素含量最豐富。因此以煮熟之甜蕎麥、苦蕎麥及苦蕎配方粉末取代小鼠基本飼料AIN-76配方,餵食以雞卵白蛋白致敏之氣喘模式小鼠六週,結果顯示苦蕎麥組和苦蕎配方組可顯著抑制發炎細胞侵潤肺部,減少血清中非特異性IgE、IgM與OVA-特異性IgG1抗體,降低以Concanavalin A (ConA)刺激脾臟細胞的Th2/Th1細胞激素比例,增加以脂多糖(lipopolysaccharide, LPS)刺激腹腔細胞的抗發炎及促發炎細胞激素的比例,此結果顯示苦蕎麥可使氣喘小鼠體內免疫平衡傾向Th1免疫反應,改善氣喘過敏反應的發生。綜合本實驗結果,證實苦蕎麥製品具有調節小鼠過敏性氣喘發炎反應的功能,推測苦蕎麥中的槲皮素成分應為降低氣喘模式小鼠過敏反應的主要活性成份。

English Abstract

Allergic asthma is classified as one of type I hypersensitivity diseases, which induces Th2-skewed immune responses. Bitter buckwheat is an emerging crop in Taiwan, which has been discovered many pharmacological functions in previous studies, including antioxidant, hypoglycemic and hypolipidemic effects as well as preventing constipation. However, the effect of bitter buckwheat on allergic asthma has not yet been investigated. Therefore, the aim of this study was to investigate the immunomodulatory effect of bitter buckwheat cultivated in Taiwan using an Ovalumin (OVA)-sensitized and -challenged BALB/c mouse model. Both in vitro and in vivo studies were included in this study. The approximate compositions and the flavonoids components of heated buckwheat powder were analyzed first. The results showed that water extracts of bitter buckwheat and common buckwheat at low concentrations exerted anti-inflammatory effects in vitro. Further analysis between flavonoids levels in buckwheat and cytokine secretions by primary cells indicated the quercetin component might be the major contributor to stimulate cytokine secretions. Furthermore, heated bitter buckwheat had the highest quercetin level among samples. Therefore, the mouse basic feed formula (AIN-76) were replaced respectively with heated common buckwheat, bitter buckwheat and bitter buckwheat formula powder to feed OVA-sensitized and -challenged asthmatic mice for six weeks. The results showed that bitter buckwheat and bitter buckwheat formula feeding suppressed on infiltration of inflammatory cells into the lungs, markedly decreased non-specific IgE, IgM and OVA-specific IgG1 antibody in sera, reduced secretion ratios of Th2/Th1cytokines by Concanavalin A (ConA)-stimulated splenocytes and increased secretion ratios of anti-/pro-inflammatory cytokines by lipopolysaccharide (LPS)-stimulated peritoneal cells. These results suggest that bitter buckwheat administration may alleviate allergic airway inflammation via its Th1-inclination and anti-inflammatory properties. In conclusion, bitter buckwheat and its products may alleviate allergic asthmatic inflammation in OVA-sensitized and -challenged asthmatic mice. Quercetin component in bitter buckwheat may be the main active ingredient for reducing allergic asthmatic reations.
