Chinese Abstract

土肉桂(Cinnamomum osmophloeum Kaneh.)為台灣固有的闊葉樹種。本研究係探討土肉桂根部之化學成分,以乙醇浸泡之萃取物,分別以正己烷、乙酸乙酯、正丁醇與水進行多次分配萃取。將乙酸乙酯層及正丁醇層以矽膠管柱層析、矽膠薄層分析及高效液相層析儀等方法做進一步的分離純化。單離後的純化合物,在測得紅外線光譜、氫核磁共振光譜、碳核磁共振光譜與質譜後,經推測為已知化合物,則與文獻比對確認其結構。若為新化合物,則需經二維核磁共振光譜及高解析質譜加以輔助,來確認其結構。 本研究共分離鑑定得到24個化合物,分別為:木酚素類(Lignans)化合物9個、黃酮類(Flavonoids)化合物9個、苯丙素類(Phenylpropanoids)化合物4個以及其他類(Others)化合物2個。其中木酚素類化合物9,9’-di-O-feruloyl-(+)-5,5’-dimethoxy- secoisolariciresinol為新化合物。 在定性分析方面,採用高效液相層析儀分析,取根部分離出之十個成分,包含:桂皮醛、桂皮酸、p-hydroxybenzoic acid 、lyoniside、trans-dihydroquercetin、sringaresinol、yangabin 、trans-3,4,5-trimethoxy cinnamic alcohol 、4’-hydroxy-5,7,3’-trimethoxyflavan-3-ol、9,9’-di-O-feruloyl-(+)-5,5’- dimethoxy-secoisolariciresinol等,進行定性分析研究。

English Abstract

Cinnamomum osmophloeum Kaneh. is an endemic tree species in Taiwan. In this study, the ethanol extract from the root of Cinnamomum osmophloeum Kaneh. was concentrated to give a residue which was added water and partitioned with ethyl acetate and n-BuOH respectively. The ethyl acetate and n-BuOH layers were separated by column chromatography on silica gel, thin layer chromatography and high performance liquid chromatography (HPLC) to give twenty-four pure compounds. The known compounds were identified by spectral analysis and comparison with those reputed in literatures. The structures of new compounds were elucidated by their 2D-NMR and HR-MS spectra. They included nine lignans, nine flavonoids, four phenylpropanoids, and two others. Among them, 9,9’-di-O-feruloyl-(+)-5,5’-dimethoxy- secoisolariciresinol (L1) is a new compound and for the first time isolated from nature. For the qualitative analysis of the chemical components in the root of C. osmophloeum, HPLC/UV chromatograms were performed and ten components isolated were used as markers. They are trans-cinnamaldehyde、trans-cinnamic acid、p-hydroxybenzoic acid、lyoniside、trans-dihydroquercetin、syringaresinol、yangabin、trans-3,4,5-trimethoxycinnamic alcohol 、and 4’-hydroxy-5,7,3’-tri- methoxyflavan-3-ol and 9,9’-di-O-feruloyl-(+)-5,5’-dimethoxy-secoisolariciresinol.
