土肉桂( Cinnamomum osmophloeum Kanehira 俗稱Indigenous cinnamon ),為台灣特有的本土闊葉樹種,本研究將土肉桂葉部進行抽取純化,進行成分之單離與鑑定。首先,將葉部以乙醇浸泡抽取,過濾後減壓濃縮得到的粗抽物以正己烷、乙酸乙酯、正丁醇與水進行分配萃取,得到各粗分配層。利用矽膠管柱色層分析法、高效能液相層析法、膠體過濾法及再結晶等方法分離純化共分得到20個純化合物,從土肉桂葉部乙酸乙酯層中除分離得到幾種常見的化合物如:包括葉綠素;二萜醇與醛類化合物;還有長鏈脂肪酸及類固醇類等化合物外,並分離得到5個黃酮苷類化合物F1、F2、F3、F6和F7以及5個苯丙素類化合物包括:桂皮醛、肉桂醇、乙酸桂皮酯、肉桂酸、香豆素等;而在正丁醇層部分則得到2個黃酮苷類的化合物F4、F5,其中於乙酸乙酯層所得到之2個黃酮苷類化合物kaempferol 3-O -(4’’-E-p-coumaroyl-α-L-rhamnopyranoside) 和kaempferol 3-O-(2’’4’’-di-E-p-coumaroyl-α-L-rhamnopyranoside)為文獻上未曾自土肉桂分離出之acylated kaempferol glycoside骨架化合物,以上各化合物之結構均依各種光譜數據資料比對而確定。 在定量方面選取從葉部中分離出的5個主要成分,包括:桂皮醛、肉桂醇、乙酸桂皮酯、Kaempferitrin與Kaempferol 3-O-α-L-rhamnopyranoside,進行定量分析研究。研究結果,發現土肉桂之葉部主要含有黃酮苷、苯丙素類化合物,其中以桂皮醛含量最多,佔乾葉含量比為1.38%。在生理活性測試方面,化合物(F7) Kaempferol 3-O-(2’’4’’-di-E-p-coumaroyl-α-L-rhamnopyranoside)對人類乳癌腫瘤細胞株(MCF-7)具有顯著的毒殺效果,其半致死濃度(IC50)為6.81μM。

English Abstract

Indigenous cinnamon (Cinnamomum osmophloeum Kanehira) (Lauraceae) is an endemic tree that grows in Taiwan’s natural hardwood forest. In this research, bioactive compounds were isolated and identified from the leaves of C. osmophloeum. The methanol extract of C. osmphloeum, was portioned successively with n-hexane, ethyl acetate, n-BuOH, and H2O, and a total of twenty compounds were isolated from these fractions of this plant using column chromatography, high performance liquid chromatography (HPLC), gel filtration and recrystallization methods, etc. Compounds including two chlorophylls, two phyto-diterpenoids alcohol, an aldehyde, a fatty acid, steroids and five phenylpropanoid, together with five flavonoid glycosides (F1, F2, F3, F6, and F7), were isolated from the ethyl acetate fraction of the leaves of C. osmophloeum. In addition, two flavonoid glycoside, F4 and F5, were isolated from the n-butanol layer. Among these isolates, kaempferol 3-O -(4’’-E-p-coumaroyl-α-L-rhamnopyranoside) and kaempferol 3-O-(2’’4’’-di-E-p -coumaroyl-α-L-rhamnopyranoside), which were the acylated kaempferol glycoside type structure, were first isolated from this plant. The structure of the above compounds was elucidated on the basis of spectroscopic analysis. Among the above compounds, the biological activity showed that compound (F7) kaempferol 3-O-(2’’4’’-di-E-p-coumaroyl-α-L-rhamnopyranoside) exhibited a strong cytotoxicity (IC50= 6.81 μM) against human breast adenocarcinoma cell line (MCF-7 cell line). Finally, we have also proceeded the quantitative analysis of five main components including trans-cinnamaldehyde, cinnamic alcohol, cinnamyl acetate, kaempferitrin, and kaempferol 3-O-α-L-rhamnopyranoside from the methanol extract of the dry leaves of C. osmphloeum using HPLC/UV. Our results demonstrate that the content of trans-cinnamaldehyde were the most abundant (1.38 %).
