以破囊壺菌Aurantiochytrium limacinum SR21醱酵生產二十二碳六烯酸(DHA)之研究

摘要


近年來,Aurantiochytrium limacinum SR21因具高油脂含量與高docosahexaenoic acid (DHA, C22:6) 產量而受到重視。本研究利用間歇性饋氧方式以維持溶氧平均值於50 %,並產生一溶氧震盪的醱酵環境,促使Aurantiochytrium limacinum SR21同時進行生長與油脂累積生成。實驗結果發現,碳氮比1.25與饋料濃度為Glycerol 100 g L-1、Yeast extract 40 g L-1, Peptone 40 g L-1的情況下,Aurantiochytrium limacinum SR21最高菌體濃度可達61.76 g L-1,油脂含量最高65.2%、DHA濃度20.3 g L-1、DHA產率122.62 mg L-1 h-1。溶氧震盪的醱酵環境可以改變Aurantiochytrium limacinum SR21SR21胞內脂肪酸組成。在醱酵中後期,Aurantiochytrium limacinum SR21胞內的飽和脂肪酸C16:0比例大幅降低至5 %以下,並提高不飽和脂肪酸C22:6的比例至70 %。 藉由添加Oleic Acid並結合溶氧震盪的醱酵環境,可促使Aurantiochytrium limacinum SR21將油酸轉化合成DHA。實驗結果發現,以Oleic Acid 50 g L-1進行醱酵培養,Aurantiochytrium limacinum SR21可獲得最高菌體濃度可達44.87 g L-1,油脂含量最高95.4 %、DHA濃度23.54 g L-1、DHA產率367.79 mg L-1 h-1。此實驗結果可以大幅降低DHA原料成本,且大幅提升DHA產率。
