現今的時代,永續綠能工業已經是目前化工業的主要研究方向,在此研究領域中酵素法以其低能耗與催化複雜分子的表現受到注目。最大的問題在於酵素價格昂貴,需要開發固定化技術。為了解決這些問題,本實驗利用種植完的台灣菇類太空包(食用菇固態廢棄培養基),利用三相分離方法成功萃取高濃度含有漆酶(laccase)之產物。再將其以光催化一鍋法固定化於(NMBA)粒子上,作為酵素薄膜反應器處理染料廢水。 本實驗使用兩種染料,Acid Blue 25和Acid Orange 7。活性測試則以BCA以及ABTS。以杏鮑菇(Pleurotus P. eryngii)和秀珍菇(Pleurotus P. ostreatus)的萃取物和商業用酵素作為對照組。本實驗以N, N'-亞甲基二丙烯醯胺(N, N'-methylenebisacrylamide, NMBA)成功造粒,商業酵素處理效果最佳(約75%),Acid Blue 25和Acid Orange 7兩種染料上都有良好的表現。 所使用兩種萃取的漆酶雖然濃度上差異不大,但是用ABTS檢測的結果,杏鮑菇的表現比秀珍菇來的優秀。所萃取的漆酶表現上雖然不及商業用酵素,但也具有商業用酵的60%。

English Abstract

Today, the Green technology has been the main focus of the chemical industry. The enzyme catalysis has attracted attention for its low energy consumption and capability of synthesis of complex molecules. In this experiment, the solid waste culture medium of edible mushrooms was used to produce extracts containing laccase (E.C.) by three-phase partitioning. The extracts can be used to degrade synthetic dyes in the waste water. In this study, two dyes, Acid Blue 25 and Acid Orange 7, served as degrading substrates.. After the one pot photosynthesis with NMBA, extracts, the solid components in the reaction solution could be separated by filtration. This matter was presumed to be the product of the radical vinyl polymerization reaction of NMBA, which may be used as a carrier for immobilizing enzymes. Next, this carrier was applied to decolorize dyes, to good effects with commercial enzyme (75%). In contrast, extracts from cultivation medium of Pleurotus eryngii achieved 50% removal of dyes. The solid catalyst synthesized by the one-pot photosynthesis for decolorization did not only have good activity, but also was proved to be reusable in a batch reactor after three repeats of 8 hours each, while 90% of the activity was still maintained in the end. In summary, this one-pot synthesis method is expected to be used as a ubiquitous enzyme immobilization method and used to develop new solid-liquid separation based membrane enzyme reactors.
