野薑花,學名Hedychium coronarium Koenig,薑科(Zingiberaceae),薑花屬(Hedychium)。野薑花植株獨特香氣常被做為芳香精油使用。薑花屬植物有豐富的倍半萜來源,而野薑花萃取出之二萜類被廣泛其生物活性的研究。實驗分二部份進行,第一部位分別萃討野薑花葉、根、莖及花朵部的蒸餾水與甲醇萃取物之總酚含量、還原力以及DPPH自由基清除能力之關係。實驗結果顯示,三種測試以水萃取物最佳;總酚含量以葉為最高,達73.5±1.5 µg GAEs/mg extract;還原力部份葉子萃取物相對最佳,達82.9±0.3 µg Vit.C/mg extract;DPPH自由基清除能力水萃取物以花最佳,達67.35±0.01%。第二部份使用蒸餾水萃取野薑花之葉片,以半製備型HPLC分為四區進行收集。取純化物H.C.04進入NMR、IR以及MS測定結構與分子量,得分子量約為710。另取純化物H.C.03與H.C.04以PPAR kit進行測試。實驗結果發現,野薑花純化樣品H.C.03與H.C.04在濃度低下對於激發體內PPAR並無明顯效果。
Hedychium coronarium Koenig is a monocotyledonous plant which belongs to the family of Zingiberaceae. The rhizome of H. coronarium is commonly used as the raw material to extract aromatic herbal oil. Hedychium plants also contain high level of sesquiterpene. The labdane diterpenes extracted from Hedychium coronarium have been reported to be vastly used in researches for their biological activities. In this work, the determination of total phenolic content, reducing power assays, as well as DPPH radical scavenging activity assay from different parts of Hedychium coronarium such as leaves, roots, stems, and flowers were investigated. Experimental results showed that distilled water extracts presented the best outcome: The highest total phenolic content was 73.51.5 μg GAEs/mg extract, which was obtained from leaves extracts, the best reduction ability came from leaves extracts, up to 82.90.3 μg Vit.C/mg extract; and flower extracts showed the highest DPPH radical scavenging ability, which reached 67.350.01%. Furthermore, a semi-preparative HPLC was used to isolated the constituents from Hedychium coronarium leaves extracts. Identification of compound structure was achieved by taking fraction (H.C.04) from NMR, FT-IR, and MS spectrum. The major component with molecular weight of 720 was determined. Additionally, the reduction in blood glucose level was conducted by applying a PPAR kit, and results showed that no significant difference between fractions H.C.03 and H.C.04 of Hedychium coronarium extracts.
