本研究針對菇蕈多醣體誘導吳郭魚肝臟細胞增生之影響,實驗方法以體外實驗與體內實驗兩部分進行探討。體外實驗結果顯示,吳郭魚肝臟細胞經過菇蕈多醣體處理後,顯著提升細胞活性與細胞增生率。於體內實驗中,首先建立魚隻部分肝臟切除手術模式,刺激吳郭魚肝臟之增生反應,再進一步探討魚隻經過菇蕈多醣體浸泡後,是否能促進肝臟之增生以及對於肝臟組織增生相關基因表現量之影響。免疫組織化學染色法結果顯示,浸泡多醣體組之肝臟新生細胞數量顯著提升,而肝臟再生相關之細胞激素、生長因子 (TNF-α、IL-8、IGF-1、GH、PGRN、FGF20-α、FGF20-β) 基因表現量亦具有顯著差異表現,推測菇蕈多醣體藉由調控相關之細胞激素、生長因子等,有效促進吳郭魚肝臟細胞之增生。
The present study aims to examine the effects of mushroom β-glucan in vitro and in vivo on the liver regeneration using tilapia (Oreochromis niloticus) following partial hepatectomy. The results showed that when tilapia liver were treated with mushroom β-glucan the hepatocytes were significantly enhanced in terms of cellular survival and cell proliferation experiment in vitro. The results of in vivo test using the tilapia partial hepatectomy model showed that mushroom β-glucan may significantly induce liver compensatory growth. The relative gene expression for liver regeneration was also enhanced. Immunochemistry observation showed that when the experimental fish was treated with mushroom β-glucan the number of hepatocyte significantly increased, while the gene expression of the cytokines and growth factor were also significantly enhanced. It is therefore suggested that mushroom β-glucan is capable to regulate the immune response and cellular repair factors for the liver.
