Determination of Xanthine Bronchodilators in Adulterated Chinese Herbal Preparations by High Performance Liquid Chromatography

Five xanthine bronchodilators, caffeine, diprophylline, proxyphylline, theobromine and theophylline, were simultaneously assayed in traditional Chinese medicine by high performance liquid chromatography on a Merck RP-select B column. The chromatographic method was carried out using isocratic elution with methanol and 1% acetic acid (15:85, v/v). 8-Chiorotheophylilne was used as an internal standard and detection was at 270 nm. Calibration curves of the five adulterants were constructed in the range 8-160 μg/mL for theobromine and 10-200μg/mL for the others. Their correlation coefficients ranged from 0.99993 to 0.99997. Extracts were treated by Supelclean C-18 before HPLC analysis. The recoveries of the adulterants in three traditional Chinese medicinal prescriptions were also studied. Diprophylline and/or theophylline were found and assayed from traditional Chinese medicines by combined solid-phase extraction and HPLC.
