台灣原生藥用植物雞鵤刺微體繁殖與其染色體數之研究

Micropropagation and Chromosome Number of Cirsium brevicaule A. Gray-A Medicinal Plant Native in Taiwan

摘要


台灣原生藥用植物雞鵤刺微體繁殖和觀察染色體數之研究,結果如下:切取無菌苗葉片和莖節為培植體,培養於含有0.2mgL^(-1)BA+0.1mgL^(-1)NAA之MS培養基,1個月後照光處理者可獲得最大量增殖的芽體(葉片7.5個、莖節7.5個)。將初代培養所得之叢生芽體切取生長正常,帶2-3片葉片之單株,接種在添加0.1mgL^(-1)NAA之MS培養基最佳,平均根數為11.7條、平均根長4.0cm、平均莖長6.3cm及平均發根率100%。發根後之瓶苗可直接移植栽培並可得到生長良好之植株。觀察野生及組培再生植株所獲得之根端染色體數,兩者均為2n=34。

The micropropagation method and chromosome number of Cirsium brevicaule A. Gray was studied: Leaf or stem explants taken from seedlings in vitro. The highest number of shoots (7.5 or 7.5) are harvested from explants cultured on MS + 0.2 mg L^(-1) BA + 0.1 mg L^(-1) NAA under light condition after one month cultivation. The individual shoot regenerated with 2-3 leaves was on the MS medium containing 0.1 mg L^(-1) NAA for rooting. After one month, rooting shoot were 6.3 cm in length with 11.7 roots of 4.0 cm in length. After transplanting in all plantlets ex vitro grew well. Both root cell chromosome number from field and in vitro were 2n=34.
