本研究利用微體繁殖方法建立闊葉大豆(金門一條根)藥用植物之增殖系統,試驗過程包含無菌苗之建立、B5及MS培養基對芽體增殖之比較試驗、植物生長調節劑對芽體增殖及發根試驗之比較試驗。為了解不同培養基對芽體增殖之影響,利用B5與MS培養基培養闊葉大豆的培植體,結果顯示B5培養基之培植體在培養12週以後,相對於生長在MS培養基之培植體,植株較高大,且叢生芽數較多,葉片呈深綠色。另在B5培養基中添加0.4ppm的BA(Benzyl adenine)、0.04ppm的NAA(1-Naphthaleneacetic acid),培植體在培養12週後,植株生長繁茂,叢生芽體數多,且葉片呈深綠色;顯示B5培養基和添加BA和NAA對於闊葉大豆芽體增殖效果較佳。另外闊葉大豆芽體增殖試驗是以植物生長調節劑和基、中、頂三節位兩因子處理,結果顯示以添加0.1/0.01 ppm BA/NAA和0.2/0.02 ppm BA/NAA之B5培養基,培養12週後可誘導出較多的芽,平均增殖芽數為8.3個,且以基部節點作為材料誘導效果最佳,誘導產生之芽體數最多。而培植體發根實驗結果顯示為B5固態培養基中若添加0.4ppmIBA(Indole butyric acid)或NAA,培養12週後發根率高達95.6-97.8%。此外添加0.4 ppm IBA於培養基中培養12週後發根數為13.8,添加0.4 ppm NAA於培養基時發根數為19.4,較其他實驗組為多。上述結果顯示添加適當的植物生長調節劑於培養過程中,對發根及芽體增殖極為重要。另將闊葉大豆馴化移植栽培於介質中,存活率皆達100%,植株生長良好,此研究結果成功利用組織培養技術建立闊葉大豆(金門一條根)微體繁殖系統。
The proliferation system of the medicinal plant of Glycine tomentella Hayata (Kinmen I-Tiao-Gung) was established by the micropropagation protocols, including establishments of sterile seedlings, inductions of shoots and adventitious roots from explants, and comparisons of B5 and MS media for shoot multiplications. Shoot multiplications test results showed that when explants grown on B5 media, plants were taller, numbers of cluster buds from explants were more, and leaves were dark green in comparsion to explants grown on MS media. Additionally, when explants grown on B5 media with 0.4 ppm BA (Benzyl adenine) and 0.04 ppm NAA (1-Naphthaleneacetic acid) for 12 weeks, plant grew more cluster buds from explants, leaves were greener, indicating B5 media with BA and NAA were more suitable for shoot multiplications. Shoot multiplications test results further demonstrated that when explants grown on B5 media with 0.1/0.01 ppm BA/NAA and with 0.2/0.02 ppm BA/NAA had more shoot numbers than other tested media. The average shoot numbers per explant were 8.3 when grown on those media. Additionally, the basal node explants had highest shoot multiplication numbers than other two types of explants, indicating the basal node explants may serve as the better material to induce shoot multiplications. When explants grown on B5 media with 0.4 ppm IBA (Indole butyric acid) or 0.4 ppm NAA for 12 weeks, root numbers per explant were more than other tested media. The rooting rates of explants grown under those two conditions were 95.6-97.8%. The root numbers per explants when grown on B5 media with 0.4 ppm IBA and with 0.4 ppm NAA were 13.8 and 19.4, respectively, which were higher than other tested conditions. These data demonstreated that additions of plant growth regulators in the media are important for shoot and adventitious root inductions of explants. The Glycine tomentella Hayata seedlings were successfully transplanted in soil and survival rates of seedlings were 100%, suggesting optimal tissue culture protocols for the Glycine tomentella Hayata sterile seedling proliferations were successful established in this study.
