摘要


"Chlamydomonas orbicularis" CPC1為台灣中油公司由南台灣分離之本土藻株,展現高油脂含量特性,最佳可達乾重的43%,戶外50公升的光生物反應器養殖試驗其平均油脂含量為20~30%。為提升CPC1戶外油脂生產效率,本研究利用紫外線突變策略進行CPC1藻種改良,提升其油脂生產能力。本研究建立320株突變藻種,利用Nile red螢光染色法進行快速篩選,經三階段培養與產油能力篩選,取得高油脂生產速率之突變株CPC1215,油脂含量最佳可達51.8%,油脂生產速率最高可達121.8 mg/L/day,相較於野生型的93.2mg/L/day,提升了30.8%。CPC1215在培養過程中,展現低黏附性與重力沉降快速之特徵。最終以50 L 光生物反應器戶外放大培養,CPC1215突變藻種之生物質量與油脂含量分別達到2.5g/L與55.4%,整體表現均較野生型提升。本研究證實"Chlamydomonas orbicularis" CPC1215具有可用海水養殖、含油脂產率高、低黏附力、可以重力沉降收集、適合戶外培養之特性,可應用於藻油生產技術,為具商業化價值之潛力藻種。

"Chlamydomonas orbicularis" CPC1 isolated from the southern coast of Taiwan by CPC Corporation, Taiwan (CPC) showed a high lipid content which is up to 43% of dry weight, and the average lipid content of 20 to 30% in outdoor cultivation by using 50 L photobioreactors. To enhance the lipid productivity of CPC1 in outdoor cultivation, CPC1 was conducted by ultraviolet induced mutation. In this study, 320 mutants of CPC1 were established and were screened rapidly by Nile red fluorescence staining. Through the three-stage culture and screening of lipid productivity, the high lipid-producing mutant, CPC1215, was obtained. CPC1215 performed the optimal lipid content of 51.8% and the highest lipid productivity of 121.8 mg/L/day which is 30.8% higher than that of CPC1 (93.2 mg/L/day). By the observation of cultured cells, CPC1215 showed low adhesion and rapid gravitational sedimentation of properties. In outdoor cultivation by using 50 L photobioreactors, the biomass and lipid content of CPC1215 were 2.5g/L and 55.4%, respectively. This study demonstrated that "Chlamydomonas orbicularis" CPC1215 would be a candidate in the commercial and industrial algal oil producer according to its high lipid productivity, low adhesion, and it is easy to be collected by gravity sedimentation under marine salinity.
