2008至2009年台灣豬場Torque Teno Virus (TTV)流行概況之探討

摘要

本試驗旨在探討Torque Teno Virus (TTV)是否存在於台灣豬場以及其在豬隻之感染概況。TTV為屬於環狀病毒科(Circoviridae)之Anellovirus屬之小型、無封套、單股環狀DNA病毒，又可區分為基因群1和2(TTV-1/2)，該病毒在豬隻疾病可能扮演重要的角色。本研究在2008-9年間共收集台灣中南部地區33家豬場6、10、16和22週齡（每年齡層各120個）豬隻共480個血清樣本進行檢測，利用聚合酶鏈鎖反應(polymerase chain reaction; PCR)和蜂巢式PCR (nested-PCR; nPCR)檢測該病毒非轉譯區(UTR)片段，而每場的陽性樣本至少擇－進行基因定序。結果顯示豬場豬隻TTV-1/2之陽性率分別為96.88%/56.25%，而在6、10、16和22週齡豬群之TTV-1/2之陽性率分別為94.2%/30.8%、97.5%/61.7%、96.7%/67.6%和99.2%/65%，而其中48個TTV-1和45個TTV-2 nPCR陽性樣本進行基因定序，發現其與標準株Sd-TTV1p (AY823990)和Sd-TTV2p (AY823991)比較，基因相似度分別介於93-100%和92.7-100%之間。由上述結果顯示，TTV-1/2皆已廣泛流行於台灣豬場。

This study was to examine whether Torque Teno Virus (TTV) existed and how prevalent it was in Taiwan’s pig farms. TTV, belongs to the Genus Anellovirus, Family Circoviridae, is a small, non-enveloped, single-stranded and circular DNA virus. TTV can be divided into geno-group 1 and 2 (TTV-1/2) and may play an important role in causing swine diseases. During this study from year 2008 to 2009, 480 serum samples (120 each from 6-, 10-, 16-, and 22-week old pigs) collected from thirty-three pig farms were examined by polymerase chain reaction (PCR) and nested-PCR (nPCR) targeting a fragment of the viral untranslated region (UTR). Genetic sequencing was also performed from at least one of the positive samples from each farm. The nPCR results for the presence of TTV-1/2 were 96.88%/56.3% positive for all 480 pigs, and the positive rates for 6-, 10-, 16- and 22-week old pigs were 94.2%/30.8%, 97.5%/61.7%, 96.7%/67.6%, and 99.2%/65% respectively. Forty-eight 1W-land forty-five TTV-2 nPCR positive samples were then analyzed by DNA sequencing. Comparisons of the DNA sequences with the reference Sd-TTV1 p (AY823990) and SdTTV2p (AY823991) showed homologies ranged from 93 to 100% and 92.7 to 100% respectively. Taken together, TTV was widespread in pig farms in Taiwan during the time studied.
