Antitumor Agents IV-Aglailic Acid, Dimethyl Terephthalate, and Phytosterols from Aglaia elliptifolia

自大葉樹蘭提取物之抗腫瘤作用

摘要


大葉樹蘭為棟科植物,有關其化學成分及其藥理學之研究,尚未見諸文獻。在臺產植物抗癌選汰實驗中,發現本植物之酒精抽取物在試管內對人鼻咽癌細胞培養具有抑制作用,而且對P388小老鼠淋巴性白血病有抗腫瘤作用。為了尋求其有效成分,本研究使用化學方法及配合藥理實驗進行此項研究工作,由大葉樹蘭根部與莖部用酒精抽取濃縮後用不同的溶媒抽取而分成六部份:(甲)石油醚可溶部,(乙)乙醚可溶部,(丙)氯仿可溶部,(丁)水可溶部,(戊)95%酒精可溶部,(己)95%酒精不溶部。由藥理實驗中以小老鼠P388白血病為指標進行抗癌試驗發現(甲)(乙)(丙)(戊)抽取部份具有抗癌作用,而(丁)及(己)則不具有抗癌作用。本論文報導有關有效石油醚抽取部以柱層層析法來進行分離成分及決定其化學結構。結果分離出三種結晶性物質。第一種結晶經熔點, 紅外線光譜, 核磁共振譜,紫外光光譜,質譜,混熔點,薄層層析等方法之分析而確定此化合物為對苯二甲酸二甲酯。第二種結晶性物質經缸外線光譜,核磁共振譜及質譜分析而推定為一種新的高分子脂肪酸,命名之為「樹蘭酸」。第三種結晶經氣相層析法之分析而確定為三種植物固醇的混合物(豆固醇,麥胚固醇,及菜子油固醇)。由小老鼠P388白血病為指標進行藥理實驗,顯示所分離的三種結晶性物質均無抗癌作用。

Aglaia elliptifolia merr, is a member of Meliaceae. To date no pharmacological or phytochemical study on this species has appeared in literature. During the routine screening of Taiwan plants for antitumor activities, a 50% alcohol extract of this plant was found to exhibit cytotoxicity against human nasopharyngeal epidermoid carcinoma in vitro (KB cell culture) and antitumor activity in BDF1 strain of mice against P386 lymphocytic leukemia (3PS)^2. From petroleum ether soluble fraction of a 95% alcohol extract, dimeillyl terephthalate (I), β-sitosterol (II), stigmasterol (III) and campesterol (IV) have been isolated and identified. A new high fatty acid has been isolated and its structure (V) determined by the m.p. and spectroscopic data. For this new compound the name aglailic acid is proposed.
