Drug-induced gingival enlargement occurs frequently in the anterior segment of gingival tissue. Except for aesthetic disfigurement, gingival enlargement could also affect pronunciation, mastication, and oral hygiene. This article highlighted the review of clinical manifestations, histological characteristics, risk factors, pathogenesis and management of the drug-induced gingival enlargement. In addition, we also presented three clinical cases associated with calcium channel blockers, immunosuppressants and antiepileptic drugs. The patients were instructed with Bass method for oral hygiene and treated with scaling/ root planing for plaque control carefully. Around two months later, the gingival inflammed appearance obtained great soft tissue healing, and even the inter-incisor space could have auto-correction at first two cases. Furthermore, we listed Chinese and English commercial name of the drugs, which frequently induce gingival enlargement, as a reference for dentists.
