大豆發酵粉緩解肌力退化及肌肉流失之探討

Lyophilized Soybean Fermentation Broth Ameliorate Cisplatin Induced Muscle Atrophy Through Myosin Heavy Chain Expression

摘要


全球平均年齡逐年增長,世界各國都將面臨人口老年化的挑戰,而肌少症(sarcopenia)更是高齡化社會的問題之一,其特徵包含骨骼肌(skeletal muscle)減少和運動表現低下,亦是提高長者跌倒及失能的危險因子之一。另外惡病質是目前與許多疾病相關的複雜代謝綜合症,其特徵也是肌肉流失,惡病質(Cachexia)被認為普通存在於各種病患中如後天性免疫缺陷症候群、慢性阻塞肺炎、多發性硬化症、慢性心臟衰竭等。本研究採用後肢單側固定法,將小鼠活動空間限制一週,使其肌肉流失,一週後恢復正常生活,以模擬肌少症或惡病質的病癥,固定的第一天就餵食大豆發酵液其凍乾粉(soybean fermentation broth and its freeze-dried powder, SBP)連續餵食兩週,結果顯示在兩週後,小鼠肌耐力以及握力相較於對照組有顯著的提升。再以小鼠纖維母細胞(mouse fibroblasts C2C12 cell line)建立順鉑(cisplatin)誘導肌肉萎縮之細胞平台,探討SBP預防肌肉萎縮之可能機制,結果發現在試驗組的肌肉細胞肌蛋白重鏈蛋白質(myosin heavy chain,MyHC)的表現量較順鉑組增加,此研究結果說明SBP可能有助於長期臥床者肌肉萎縮情況之改善。

Every country in this world is facing an older population trend. In this aging society, sarcopenia has become one of the major healthcare problems. The symptoms of sarcopenia include reduction in skeletal muscle mass and decrease in muscle performance. All these factors can increase the elderly's falls and disability. At the same time, cachexia is also another complex metabolic syndrome in the elderly and also accompanied by muscle loss. Cachexia is a common symptom occurs in various diseases such as acquired immunodeficiency syndrome, chronic pneumonia, multiple sclerosis, chronic heart failure, etc. Therefore, our aim is to used cast immobilization procedure to restrict the mice movement to simulate sarcopenia or cachexia which is caused by muscle loss effect. The mice will be immobilized for one week and return to normal condition for another week for muscle recovery. This study will observe the feeding effect of the soybean fermentation broth and its freeze-dried powder (SBP) for two continuous weeks. Results showed that the muscle endurance and grip strength of the mouse were significantly improved compared with the control group. To investigate its underlying mechanism, we used the mouse fibroblasts (C2C12) cell line with cisplatin treatment to inducing muscle atrophy and evaluate its protective effect. The Western blot results showed SBP group expressed higher muscle growth protein (myosin heavy chain, MyHC) than the cisplatin group. The results of this study show that SBP may help to improve muscle atrophy for the long-term care patients.
