發酵乳桿菌GKF3可加強小鼠在禁錮壓力下的抗憂鬱與抗發炎反應

Anti-depression and Anti-inflammation Effects of the Probiotic Limosilactobacillus fermentum GKF3 in Mice with Restraint Stress

摘要


憂鬱症是一種情感性精神疾病,係因腦神經透過神經傳導物質包括多巴胺(dopamine)、血清素(serotonin)、正腎上腺素(norepinephrine)發生改變所引起的。先前研究指出腸道微生物可影響腦部神經精神的各種活動。本研究調查發酵乳桿菌GKF3(Limosilactobacillus fermentum GKF3)是否可改善6週齡ICR品系雄性小鼠受到禁錮壓力(restraint stress, RS)所引起的憂鬱症,試驗時,RS鼠餵食16.7 mg/kg b.w./day之活菌或熱致死死的GKF3共28天,結果顯示活菌或熱致死死的GKF3均可改善小鼠因禁錮壓力所引起的憂鬱行為,生化分析顯示腦中多巴胺及血清素顯著地增加(p<0.01),並且,促炎細胞激素介白素-6(IL-6)及腫瘤壞死因子-α(TNF-α)含量皆顯著低於RS組(p<0.001)。此結果證實活菌或熱致死的發酵乳桿菌GKF3皆可預防壓力所致的憂鬱症,且可降低發炎反應,尤其熱致死死菌GKF3具有被開發成益生菌配方的潛力。

Depression is a mood disorder due to imbalanced levels of dopamine, serotonin, or norepinephrine in the brain. Studies have shown that intestinal microbes can affect neuropsychological activities. In this study, we investigated whether the probiotic Limosilactobacillus fermentum GKF3 has any beneficial effect on depression using 6-week-old male ICR mice with depression induced by restraint stress (RS). These mice were fed with live or heat-killed GKF3 at a dose of 16.7 mg/kg b.w. per day for 28 days. Results showed that both live and heat-killed GKF3 were effective in alleviating RS-induced depression in mice. Biochemical analyses revealed that the levels of dopamine and serotonin serotonin in the brain were significantly increased (p<0.01), and serum levels of pro-inflammatory cytokines IL-6 and TNF-α were significantly decreased in RS (p<0.001). These results suggest that both live and heat-killed L. fermentum GKF3 are effective in preventing stress related depression and that heat-killed GKF3 can potenti ally be developed to become a good probiotic formulation.
