停留時間對抗體-抗原解離力與丙胺酸去摺疊力之影響及酸鹼值對丙胺酸摺疊程度之探討

摘要


本文是以原子力顯微鏡探討單一生物分子之間作用力為主,內容分成一量測Anti-Human IgG1與Human IgG1之間的解離力,探討解離力與壓入深度以及停留時間的關係,比較下壓時間與停留時間對解離力的影響,從數據中知道到了停留時間十秒時仍未有穩定的解離力出現,可見構形仍然在持續改變著。另外研究蛋白質拉伸的實驗,實驗樣本為丙胺酸(poly-alanine),探討去摺疊力與實驗環境酸鹼度以及停留時間的關係,並將實驗所得的力-距離曲線與Worm Like Chain model做結合,如此可以進而求得丙胺酸的抗彎長度(persistence length)與實驗環境酸鹼度的關係,並與圓旋光光譜儀的實驗結果最比較,實驗結果證實不同酸鹼溶液下去摺疊力的大小與該胺基酸的pKa值息息相關。
