鏈球菌全細胞疫苗在烏魚保護效力之評估

Evaluation of the Protection of Streptococcal Whole-cell Vaccine on Grey Mullet (Mugil cephalus)

摘要


自台灣的虹鱒、烏魚與吳郭魚等分離出的三種鏈球菌,經16s rDNA定序分析加以確認菌種分別為:Enterococcus feacalis、Streptococcus、Lactococcus gravieae。將此三株鏈球菌以不同組合方式製備成福馬林殺滅全細胞疫苗,以高滲透壓浸泡免疫方式對烏魚(Mugil cephalus)進行免疫,並評估此等疫苗的相對存活率(relative percent survival; RPS)。免疫攻毒後之相對存活率為58.3-73.2%,血清抗體力價可在免疫後第二週檢測而得,而到第三至四週有明顯上升。烏魚免疫後一週進行攻毒,其入侵的鏈球菌在第三天後便無法分離到。本實驗製備的福馬林殺滅全細胞疫苗可以誘發保護效力清除早期數週入侵魚體之鏈球菌,即便免疫後早期的血清抗體力價仍然低。

Three species of streptococcaceae ( Enterococcus feacalis, Streptococcus, and Lactococcus gravieae ) isolated from cultured rainbow trout, mullet and tilapia of Taiwan were identified by 16s rDNA sequencing. The formalin-inactivated whole cell vaccines of these streptococci were prepared. Grey Mullet (Mugil cephalus) were vaccinated by hyperosmotic immersion to evaluate the relative percent survival (RPS) of these vaccines. Besides, serum antibody titer of vaccinated fish has also been detected. The RPS was 58.3-73.2%, and the antibody titer can be detected at 2(superscript nd) week and then significantly raised during 3(superscript th) and 4(superscript th) week after vaccinated. These formalin-inactivated vaccines of streptococci elicited a protective response to clear off the invasive streptococci in fishes during early weeks, even though the antibody titer were still low.
