Detection of Specific Serum IgE in Dog with Allergic Dermatitis and the Identification of Major Brewer's Yeast Allergens

過敏性皮膚炎犬隻血清之特異性IgE偵測及啤酒酵母主要致過敏原成分之鑑定

摘要


為決定具有過敏臨床症狀的台灣家犬血清過敏測試的陽性率以及常見的過敏原,並鑑定啤酒酵母(Saccharomyces cerevisiae)的主要致過敏原。採集全台2006至2008年,505隻疑似異位性皮膚炎的家犬血清樣本,利用蛋白質微陣列分析血清裡對抗62種不同過敏原的特異性IgE,並挑選出對S. cerevisiae反應陽性的血清,進一步使用西方墨點法分析S. cerevisiae中之致過敏原。血清樣本中80.20%(405/505)含有至少一種對抗過敏原的特異性IgE,其中29.50%的陽性樣本含有對抗歐洲室塵(Dermatophagoides pteronyssinus)的特異性IgE,僅17.03%含有對抗美洲室塵(D. farina)的特異性IgE,兩相比較有極顯著差異(P<0.0001)。在季節方面,夏季採集的樣本(76.15%)與冬季(53.68%)的陽性率相較在統計學上有顯著的差異(P=0.0084),春天(73.62%)與冬天的陽性率相較亦有顯著差異(P=0.0242)。在犬隻年齡的部分,於特異性IgE部分並未發現顯著差異(P=0.97)。啤酒酵母中的主要致過敏蛋白為磷酸丙糖異構酶(triosephosphate isomerase)、磷酸甘油酸變位(phosphoglycerate mutase)、烯醇酶(enolase)、α葡萄糖苷酶(alpha-glucosidase)。推測塵蟎為台灣犬隻最普遍的過敏原。此外如果在低過敏原的飼料中添加了啤酒酵母可能使異位性皮膚炎的症狀更嚴重。

To determine the positive rate of serum allergy test and the common allergens obtained from household dogs with allergic symptoms in Taiwan, and to identify major allergens in brewers yeast (Saccharomyces cerevisiae), sera from 505 household dogs suspected with canine atopic dermatitis were collected between 2006-2008. Protein microarray of 505 sera from dogs was used to identify specific IgE against 62 different allergens, and western blot analysis was further carried out for dogs tested positive for S. cerevisiae. The results showed that 405/505 (80.20%) of the household dogs had serum IgE against at least 1 or more allergens tested. A greater percentage of the household dog sera had IgE against Dermatophagoides pteronyssinus (29.50%) proteins than against Dermatophagoides farina (17.03%; P<0.0001). The study demonstrated a significant difference between the positive rate obtained in summer (76.15%) and winter (53.68%; P=0.0084); also in spring (71.62%) and winter (P=0.0242). Age did not play a significant role in the outcome of sIgE test (P=0.97). The major proteins identified in S. cerevisiae were triosephosphate isomerase, phosphoglycerate mutase, enolase and alpha-glucosidase. Dogs suspected with allergy in Taiwan are most commonly tested positive for mites. The addition of brewers yeast in hypoallergenic diets may causes flares of atopic dermatitis symptoms.
