苦瓜植物生長素受體基因之選殖與分析

Cloning and Analysis of Auxin Receptor Gene from Momordica charantia

摘要


爲了瞭解植物生長素對苦瓜生長發育的調控機制,本研究以阿拉伯芥植物生長素受體AtTIR1基因片段爲探針,篩選苦瓜cDNA庫,得到含有苦瓜植物生長素受體McTIR1 cDNA的選殖系。進一步以McTIR1 cDNA爲探針篩選苦瓜基因組庫,得到包含苦瓜McTIR1基因的基因組選殖系λMcAR7。McTIR1具3個顯子及2個隱子,編碼594個胺基酸,其上具有F-box及白胺酸重複區之保守序列,預測之分子量爲66.8 kDa,等電點爲6.27,與阿拉伯芥及棉花TIR1之胺基酸序列同源性分別爲78%與77.2%。南方氏雜交分析結果顯示McTIR1爲單拷貝基因。

For elucidation of the molecular mechanism of auxin regulation on growth and development in bitter gourd, a cDNA encoding auxin receptor McTIR1 was firstly isolated from a cDNA library of bitter gourd using Arabidopsis AtTIR1 as probe. The McTIR1 cDNA was then used to screen the genomic library of bitter gourd. The genomic clone λMcAR7 contained McTIR1 gene with 3 exons and 2 introns. McTIR1 encodes an open reading frame consisting of 594 amino acids and F-box motif and leucine-rich repeats are found. This deduced polypeptide, designated as McTIR1, is with a calculated molecular weight of 66.8 kDa and a predicted pI of 6.27. The McTIR1 protein exhibits 78.0% and 77.2% identity in amino acid sequences with TIR1 from Arabidopsis thaliana and Gossypium hirsutum, respectively. Southern analysis indicated that McTIR1 is a single-copy gene.
