九重葛黃化葉脈鑲帶病毒全長度基因序列之分析

摘要


於高雄市大樹區佛光山發現九重葛(Bougainvillea spectabilis)植株矮化和生長不良的病株,其葉片褪綠,沿葉脈出現黃化鑲帶(chlorotic vein-banding),及葉片捲曲等病徵。本研究取罹病葉片進行病毒純化。病毒形態呈桿菌狀(bacilliform),大小約為120-130×30nm。病毒具單片段基因體DNA,大小約8.7 kb。基因序列解析顯示其為九重葛黃化葉脈鑲帶病毒 (Bougainvillea chlorotic vein banding virus, BCVBV)。病毒之全長度基因組(accession number: EU034539)含8,759個核苷酸,具四個開放轉譯架構(ORFs 1, 2, 3, 和4)。BCVBV與12種Badnavirus屬病毒之全長度基因核苷酸序列相同度普遍偏低(36 - 45%)。

Bougainvillea (Bougainvillea spectabilis) with symptoms of stunting and decline was discovered at Fuo Guang Shan in Kaoshiong. The affected leaves showed mottling, chlorosis, chlorotic vein banding, and leaf distortion. A virus was purified from diseased leaves and found to contain bacilliform virus particles of about 130 x 30 nm in size. The virus contains a monopartite genome of 8,759 nucleotides (accession number EU034539) and was identified as Bougainvillea chlorotic vein banding virus (BCVBV). The viral genome encodes 4 open reading frames (ORFs 1, 2, 3, and 4). The full length genome sequence of BCVBV displays relatively low nucleotide identities (36 - 45%) with those of 12 species of the Badnavirus.
