近紅外光分析儀(NIRS)快速測定狼尾草之粗蛋白質及水溶性碳水化合物

Quick determination of the crude protein and water soluble carbohydrate of napiergrass by near infrared reflectance spectroscopy (NIRS)

摘要


本試驗目的在建立利用近紅外光分析儀(near infrared reflectance spectroscopy, NIRS)快速測定狼尾草(Pennisetum purpureum)植物體之粗蛋白質(crude protein, CP) 及水溶性碳水化合物(water soluble carbohydrate, WSC) 含量之檢量線(calibration equation)。試驗前收集來源不同的樣品,狼尾草品種包括狼尾草台畜草一、二、三、四及五號,和不同年代、割期、季節、種植地及不同肥料處理等1000 多個樣品。將每個樣品作NIRS 掃描分析,並同時進行傳統的化學分析。採用淨最小平方差迴歸(partial least squares regression, PLSR)建立狼尾草CP 及WSC 快速測定檢量線。試驗結果顯示,利用NIRS 建立狼尾草CP 及WSC 之檢量線與化學分析方法之決定係數(coefficient of determination, R^2)分別為0.96 及0.94。檢量標準偏差(standard error of calibration, SEC)分別為0.62 及0.73。交互驗證標準偏差(standard error of cross validation, SECV)分別為0.68 及0.78。交互驗證決定係數(coefficient of determination for cross validation, 1-VR)分別為0.95及0.93。NIRS 檢量線測試結果,狼尾草CP 及WSC 之實測平均含量與NIRS 預測的平均含量之差異率(percent of difference)分別是-2.94 及6.73%。利用NIRS 檢量線測定狼尾草CP 及WSC 含量與化學分析結果之相關係數(correlation coefficient, r)分別是0.95 及0.88。NIRS 測試結果,狼尾草成分CP 及WSC 之檢量線的準確度高。本試驗結果可供酪農調配每日完全日糧外,亦可供育種學家育種研究上之應用及參考。

The objectives of this study were to establish the calibration equations for quick determination of crude protein (CP) and water soluble carbohydrate (WSC) of napiergrass (Pennisetum purpureum) by the near infrared reflectance spectroscopy (NIRS) and providing information to prepare feeding for rumen animal. The samples of napiergrass were collected from diverse sources, including different varieties of napiergrass (NP cv.TS1~TS5) with different years of cultivation, cutting stage, growing season, planted area and fertilizers applied. The samples were divided to two parts. One part of the samples was taken for determining CP and WSC by traditional chemical analysis and the other part was scanned by NIRS. The partial least squares regression (PLSR) was used to establish the calibration equations. The results showed that the coefficients of determination (R^2) between calibration equation and chemical analysis for CP and WSC of napiergrass were 0.96 and 0.94, respectively. The standard errors of calibration (SEC) were 0.62 and 0.73, respectively. The standard error of cross validation (SECV) was 0.68 and 0.78, respectively. The coefficients of cross validation (1-VR) were 0.95 and 0.93, respectively. The calibration equations of NIRS were be test and verified with another 80 new napiergrass samples for determining CP and WSC. The results showed that the percents of difference between chemical method and NIRS prediction were -2.94 and 6.73%, respectively. The correlating coefficient (r) between chemical analysis and NIRS prediction were 0.95 and 0.88 for determining CP and WSC, respectively. The results showed that the calibration equations of NIRS for predicting the CP and WSC of napiergrass could be used to quickly determine those of contents of napiergrass. The results might be suggested to dairy farmer for preparing feeding and breeders for breeding the new forage crops.
