桑葉萃取物抑制糖尿病性腦神經細胞退化疾病

摘要


近年來,糖尿病一直都是國人最常見的慢性病之一,更位居國內十大死因的第五名。糖尿病導致的高血糖及高血脂皆會使粒線體產生氧化的壓力而死亡,後期就有神經病變的情形。 本實驗室之前的研究指出,桑葉萃取物可降低粥狀動脈硬化而保護心血管的作用、減緩酒精與非酒精性肝炎的情形、減緩糖尿病性脂肪肝炎的效果。本篇所探討的是糖尿病引起的神經退化性疾病,藉由具抗氧化成分氯原酸的桑葉水萃物(MLE)餵食糖尿病基因缺陷鼠db/db mice,使用免疫螢光染色及西方墨點法發現給予高脂餵食的小鼠大腦中,紋狀體及中腦黑質部分的神經病變相關蛋白α-synuclein及β-amyloid表現都有增加的情形,而給予MLE餵食的小鼠則有減緩;在多巴胺相關蛋白TH的部分,餵食MLE的組別則有明顯的增加。另外,模擬高油與高糖的環境,培養神經母細胞瘤細胞SH-SY5Y,使用MTT試驗發現氯原酸及新氯原酸的處理可以改善SH-SY5Y細胞的死亡情形。 基於上述實驗,證實含有氯原酸及新氯原酸的MLE可藉由抗氧化抑制神經細胞的死亡,增加多巴胺相關蛋白。因此,可用於發展健康食品,改善糖尿病患者神經病變之症狀。

In recent years, diabetes has been one of the most common chronic diseases in Taiwan. It’s also the fifth among the Top Ten Causes of Death in Taiwan. Hyperglycemia, a characteristic of diabetes, and hyperlipidemia cause oxidative stress and lead the mitochondria death. Then, neuropathy may occur in later stage. Our previous researches show that mulberry leaf extract (MLE) can reduce atherosclerosis and protect cardiovascular function, ameliorate alcoholic and non-alcoholic hepatitis, and improve the diabetic fatty liver disease. In this article, we investigate the effect of MLE rich in chlorogenic acid and neochlorogenic acid on neurodegenerative diseases caused by diabetes. We used Immunohistochemistry and Western Blot to find that the expression of α-synuclein and β-amyloid increased in striatum and midbrain substantia nigra in High-Fat-Diet (HFD) group, but the MLE decreased the α-synuclein and β-amyloid levels induced by HFD. Then, the expression of TH showed a significant increase in MLE group. In addition, the protective efficacy of MLE on neuroblastoma cells SH-SY5Y in a high-oil and high-glucose environment was also evaluated. Our MTT assay results showed that chlorogenic acid and neochlorogenic acid could inhibit the death of SH-SY5Y in high-glucose and high-oil environment. Based on the experiments, we confirmed that chlorogenic acid and neochlorogenic acid of MLE can inhibit the death of nerve cells and increase dopamine-related proteins. Therefore, it has the possibility of being the healthy food to improve the symptoms of neuropathy in diabetic patients.
