菱角殼乙醇萃取物抗乳癌細胞的作用機制的探討

Studies on the Anti-Breast Cancer Mechanisms of Trapa Natans L.

摘要


自古以來菱角就被認為是營養成分很高的食物。本研究主旨在探討菱角殼抑制乳癌細胞之增生以及作用機制之探討。以MTT初步篩檢發現,菱角殼乙醇萃取物對乳癌細胞MDA-MB-231的增生抑制效果最佳,因此繼續對MDA-MB-231的作用機制進行探討並且做更深入的實驗。結果顯示:在Trypan blue試驗中,菱角殼乙醇萃取物可以明顯抑制MDA-MB-231之增生;在48小時藥物暴露下,會造成MDA-MB-231的DNA斷裂;並且增加細胞週期的Sub-G1期,及造成ROS的減少;也會使Bad的表現增加以及Bcl-2和caspase 9的表現下降。

The Trapa natans L. has been believed rich in nutritional compositions for a long time. The purpose of this study was to investigate the anti-proliferation activity and action mechanism of the ethanol extract of Trapa natans L. to MDA-MB-231 cells. Firstly, from the MTT assay the ethanol extract of Trapa natans L. showed the great anti-proliferation activity to MDA-MB-231 cells. Therefore, we focused study the action mechanism of the ethanol extract of Trapa natans to MDA-MB-231 cells. The result showed the ethanol extract of Trapa natans L. made the cell ladders of MDA-MB-231; increased the cell populations at the Sub-G1 phase; finally decreased the intracellular ROS; and increased the expression of Bad; decreased Bcl-2 and pro-caspase 9.
