臺灣杉木質部與韌皮部蛋白質之差異表現

Differential Expression between the Proteins of Xylem and Phloem in Taiwania (Taiwania Cryptomerioides)

摘要


本試驗成功地利用二維膠體電泳法解析臺灣杉木質部與韌皮部表現的蛋白質,經過比對分析後找出木質部與韌皮部差異表現的蛋白質點,以及定位出這些蛋白質點的等電點與分子量,再利用資料庫搜尋比對,定義其可能的功能以及分類,進而明瞭組織分化中蛋白質表現的差異處。在木質部方面,共找到與韌皮部差異的28個點,推測為胺基酸代謝、二次代謝物和細胞支架等相關蛋白質,其中,部分胺基酸代謝和二次代謝物相關的蛋白質與木材形成有關;在韌皮部方面則找到9個差異點,推測為碳水化合物代謝及輔因子與維他命代謝相關之蛋白質,其中,碳水化合物相關蛋白質可能與細胞壁中的多醣有關,而輔因子與維他命代謝相關之蛋白質,則可能與木本植物的韌皮部的木質素生合成有著密切的關係。

The differential expression of xylem and phloem proteins in Taiwania was successfully separated by two-dimensional (2-D) gel electrophoresis. Following by gel image analysis, mass sequence analysis, and data searching, the differential proteins spots between xylem and phloem were distinguished. Moreover, the functions of these proteins were identified and classified. In xylem, 28 differential proteins were found and presumed that they were proteins related to amino acid metabolism, secondary metabolites and cell cytoskeleton etc. A part of the amino acid metabolism and secondary metabolites related proteins might be correlated to wood formation. In phloem, we found 9 differential proteins and presumed that they were proteins related to metabolism of cofactors and vitamins and carbohydrate metabolism. The carbohydrate metabolism related proteins might be correlated to the polysaccharides in the cell wall. Besides, metabolism of cofactors and vitamins related proteins play the important role in lignin biosynthesis of phloem.
