台灣雲杉MADS-box基因之選殖及其特性分析

Cloning and Characterization of MADS-Box Gene in Picea Morrisonicola

摘要


依據已發表之植物開花相關的序列比對結果,本研究利用保守區之核酸序列設計一組引子對,以台灣雲杉毬果所萃取出的RNA做模版,進行聚合酶連鎖反應,所獲得之反轉錄cDNA片段共有856個核苷酸,其蛋白質轉譯區(coding region)具有669個核苷酸,可轉譯出222個胺基酸之蛋白質,並將其命名為PMHMADS。經過核酸及胺基酸序列的比對分析,顯示PMHMADS的序列結構和已發表之MADS-box基因,同樣具有MADS-box domain與K-domain兩個保守性高的區域。經由親緣分析的結果顯示,從台灣雲杉毬果中選殖出的PMHMADS基因,和其他木本植物的MADS-box基因具極高的相似度,其核酸相似度可達85%~99%;而胺基酸序列則可達83%~99%的相似度。由分析之結果可知,PMHMADS基因應歸類屬於AGAMOUS(AG)家族,為C type。

Based on the MADS-box gene associated with flowering from published database, a pair of primers was designed from the conserved region. After reverse-transcription polymerase chain reactions (RT-PCR) using total RNA from the cone of Picea morrisonicola as templates, 856 base pairs (bp) of cDNA fragments were obtained. The coding region of this DNA fragment was 669 base pairs, which can be translated to 222 amino acids, named PMHMADS. On the basis of DNA and protein sequence analysis, PMHMADS contains two highly conserved MADS-box domain and K-domain, which were the same as other MADS genes previously reported. Comparing PMHMADS with those of Picea abies, Picea mariana, Pinus resinosa, Pinus radiata and Ginkgo biloba, it has shown up to 85%-99% of similarity. Moreover, the amino acid sequence prediction data also presented 83%-99%of similarity. This result demonstrated that PMHMADS gene should belong to AGAMOUS gene family, which is known as C type.
