綜論:穿心蓮內酯之抗腫瘤作用及其分子機制

摘要


穿心蓮(Andrographis paniculata(Burm.F.)Nees)植株生長於亞熱帶地區,是東南亞國家常使用的一種中草藥。穿心蓮萃取物成分主要為含二帖內酯化合物(diterpene lactones),其中約有70%為穿心蓮內酯(andrographolide)。穿心蓮內酯普遍應用於許多臨床治療,包含發炎、腹瀉、發燒與感染相關疾病。過去研究指出,穿心蓮內酯具有免疫調節與抑制癌細胞生長的活性,然而其抗癌作用及其分子機制尚未被釐清。綜合我們近年來研究發現,穿心蓮內酯可以使大腸直腸癌Lovo細胞的生長週期停滯(cell cycle arrest)於G1時期,也可以抑制Lovo細胞與人類非小細胞肺癌A549癌細胞移動與侵襲(migration and invasion)作用。另外,穿心蓮內酯可以經由負調控低氧壓誘導因子1α(hypoxia-inducible factor-1alpha,HIF-1α)抑制A549癌細胞參與血管新生(angiogenesis)與惡化轉移(malignant metastasis)作用。這些發現證明穿心蓮內酯在癌症治療上具有發展為抗癌藥物的潛力。

Andrographis paniculata (Burm. F.) Nees (Acathaceae) is a traditional herbal medicine widely used in Southeast Asian countries. Its main components are diterpene lactones, and 70% of the plant extract is constituted of andrographolide (Andro). Andro has been widely used in the clinic treatment of inflammation, diarrhea, fever, and other infectious diseases. Previous studies reported that Andro possesses immunomodulatory and tumor-inhibitory activities, and therefore has the potential to be applied as a chemotherapeutic agent. However, the molecular mechanisms involved in the anticancer effect of Andro are poorly understood. Our recent studies showed that Andro was able to induce cell cycle arrest at the G1 phase of human colorectal carcinoma Lovo cells. Those studies also demonstrated that Andro can inhibit tumor migration and invasion of Lovo cells and human non-smallcell lung cancer (NSCLC) A549 cells. Furthermore, Andro can downregulate hypoxia-inducible factor (HIF)-1α to inhibit tumor angiogenesis and malignant metastasis in A549 cells. These results highlight the potential effects of Andro, which can be developed as an anticancer agent in the future.
